摘 要:目的:观察复脾汤对STZ模型大鼠血糖、LEP、INS及脂肪因子Apelin、vaspin指标的影响,探讨复脾汤治疗STZ模型大鼠的疗效,以及脂肪因子在糖尿病的作用。方法:将采用高脂、高糖饲料,造模成功的大鼠随机分成复脾汤组10只、二甲双胍组10只、模型组10只和空白组10只。造模2周后进行药物干预,模型组和空白组均用生理盐水(每1 kg体质量10 mL)灌胃;二甲双胍组采用二甲双胍悬浮液,按每1 kg体质量0.089 g灌胃;复脾汤组采用复脾汤煎剂,按每1 kg体质量14.2 g的剂量灌胃。结果:治疗后复脾汤组和二甲双胍组FBG、2hBG、14dBG和TG水平均低于模型组,复脾汤组和二甲双胍组与模型组比较,差异均有统计学意义(P<0.05)。复脾汤组与二甲双胍组之间比较,两组BG、2hBG、TG水平比较,差异无统计学意义(P>0.05),14dBG水平比较,差异有统计学意义(P<0.05)。治疗后复脾汤组和二甲双胍组INS1升高,INS2表达降低,复脾汤组和二甲双胍组与模型组比较,差异均有统计学意义(P<0.05)。复脾汤组和二甲双胍组比较,两组大鼠INS1、INS2水平变化不明显,差异无统计学意义(P>0.05)。治疗后复脾汤组和二甲双胍组LEP、Apelin、vaspin表达均降低,复脾汤组和二甲双胍组与模型组比较,差异均有统计学意义(P<0.05)。复脾汤组和二甲双胍组LEP、Apelin、vaspin表达情况比较,水平变化不明显,差异无统计学意义(P>0.05)。结论:复脾汤能调节血糖、血脂、胰岛素、脂肪因子(LEP)以及新型脂肪因子(Apelin、vaspin)水平,治疗STZ模型大鼠的疗效显著。通过本研究也证实,脂肪因子(Apelin、vaspin)在糖尿病的发生、发展,以及糖尿病抵抗等方面有重要作用。Objective:To observe the effects of Fupi Decoction(FPD)on blood sugar,LEP,INS,Apelin and vaspin in STZ model rats,and to explore the therapeutic effect of FPD on STZ model rats and the role of adipokines in diabetes mellitus(DM).Methods:After successful modeling,the rats were randomly divided into FPD group,Metformin group,the model group and the blank group.2 weeks after modeling,the model group and the blank group were given normal saline;Metformin group and FPD group were correspondingly given medication of Metformin Suspension and Fupi Decoction.Results:The values of FBG,2hBG,14dBG and TG were significantly lower in FPD group and Metformin group than those in the model group after the treatment(P<0.05);there were no statistical differences in BG,2hBG and TG between FPD group and Metformin group(P>0.05),but there was significant difference in 14dBG between the two groups(P<0.05).The expression of INS1 was significantly increased and the expression of INS2 was significantly decreased in FPD group and Metformin group compared to those in the model group after the treatment(P<0.05);of which there were no statistical differences between FPD group and Metformin group(P>0.05).The expressions of LEP,Apelin and vaspin were significantly decreased both in FPD group and Metformin group compared to those in the model group(P<0.05);of which there were no statistical differences between FPD group and Metformin group(P>0.05).Conclusion:FPD can regulate blood sugar,blood lipid,insulin,fat factor(LEP)and new type of fat factors(Apelin,vaspin),it has a significant therapeutic effect on STZ model rats.This study also confirmed that adipokines(Apelin,vaspin)play an important role in the occurrence and development of DM,as well as in the resistance to DM.
