摘  要:目的:以人气道上皮细胞16HBE为研究对象,通过体外实验探究白藜芦醇对H2O2作用下人气道上皮细胞16HBE中黏蛋白5AC(MUC5AC)表达的影响。方法:建立H2O2诱导的细胞氧化应激模型,并采用白藜芦醇进行干预,检测活性氧(ROS)和总抗氧能力(T-AOC)水平;RT-PCR法检测MUC5AC基因表达水平;Western blot法检测MUC5AC蛋白表达水平。结果:H2O2作用后16HBE细胞的ROS水平高于对照组,T-AOC水平低于对照组(P<0.05),MUC5AC mRNA水平明显高于对照组(P<0.001),MUC5AC蛋白表达水平亦高于对照组(P<0.001),给予白藜芦醇干预后,H2O2导致的上述效应明显得到抑制。结论:白藜芦醇能够抑制氧化应激减少人气道上皮细胞MUC5AC的表达。Objective:To investigate the effect of resveratrol on the expression of MUC5AC in human airway epithelial cells 16HBE under the action of H2O2 in vitro.Methods:The oxidative stress model was induced by H2O2 and the levels of ROS and T-AOC were detected after resveratrol intervention.The expression level of MUC5AC gene was detected by RT-PCR.Expression level of MUC5AC protein was detected by Western blot method.Results:After H2O2 intervention,the ROS level of 16HBE cells was higher than that of the control group,the T-AOC level was lower than that of the control group(P<0.05),the mRNA level of MUC5AC was significantly higher than that of the control group(P<0.001),and the protein expression level of MUC5AC was also higher than that of the control group(P<0.001).However,after resveratrol intervention,all above effects were inhibited significantly.Conclusion:Resveratrol can inhibit oxidative stress and reduce the expression of MUC5AC in human airway epithelial cells.
