摘　　要：目的:观察苏木乙酸乙酯提取物对慢性病毒性心肌炎小鼠心脏功能及心肌组织p38MAPK mRNA的影响,探讨其可能的作用机制。方法:采用反复增量腹腔接种CVB3建立慢性病毒性心肌炎小鼠模型。10只雄性Balb/c小鼠作为空白对照组;75只慢性病毒性心肌炎模型小鼠随机分为模型对照组、苏木乙酸乙酯提取物组(简称苏木组)、环孢素组,每组各25只。心脏超声测定小鼠心脏左室舒张末期内径(LVEDD),左室收缩末期内径(LVESD),左室射血分数(LVEF)和左室缩短分数(FS);Real-Time PCR法检测小鼠心肌组织p38MAPK mRNA的表达。结果:1)小鼠心脏功能:与空白对照组相比,模型对照组小鼠LVEDD和LVESD显著扩大,EF和FS显著下降(P<0.01);与模型对照组相比,环孢素组和苏木组的LVEDD和LVESD均明显减小,EF和FS明显增大(P<0.05);苏木组与环孢素组比较无统计学意义(P>0.05)。2)小鼠心肌组织p38MAPK mRNA表达:与空白对照组相比,模型对照组的p38MAPK mRNA表达明显增加(P<0.05);与模型对照组相比,环孢素组、苏木组的p38MAPK mRNA表达均明显降低(P<0.05);环孢素和苏木组的p38MAPK mRNA表达无明显差异(P>0.05)。结论:1)苏木乙酸乙酯提取物能够改善慢性病毒性心肌炎小鼠的心脏功能,抑制心脏肥大。2)苏木乙酸乙酯提取物能够抑制慢性病毒性心肌炎小鼠心肌组织p38MAPK mRNA的表达,是其治疗慢性病毒性心肌炎的机制之一。Objective:To observe the effect of ethyl acetate extract of Sappan on cardiac function and p38MAPK mRNA in mice with chronic viral myocarditis,and to explore its possible mechanism.Methods:The mice model of chronic viral myocarditis was established by repeated incremental intraperitoneal inoculation of CVB3.10 male Balb/c mice were taken as the blank control,and 75 chronic viral myocarditis mice were randomly divided into the model control group,Sappan group and Cyclosporin group,with 25 mice in each group.Left ventricular end diastolic diameter(LVEDD),left ventricular end systolic diameter(LVESD),left ventricular ejection fraction(LVEF)and left ventricular fraction shortening(FS)were measured by echocardiography.The expression of p38MAPK mRNA was detected by Real-Time PCR.Results:In terms of cardiac function of mice,LVEDD and LVESD were significantly increased,and EF and FS were significantly decreased in the model control group compared to those in the the blank control group(P<0.01);compared to those in the model control group,LVEDD and LVESD were significantly decreased,and EF and FS were significantly increased in Cyclosporine group and Sappan group(P<0.05);of which there were no significant differences between Sappan group and Cyclosporin group(P>0.05).In terms of the expression of p38MAPK mRNA in myocardial tissue of mice,it was significantly increased in the model control group compared to that in the blank control group(P<0.05);it was significantly decreased in Cyclosporine group and Sappan group compared to that in the model control group(P<0.05);there was no significant difference in p38MAPK mRNA expression between Cyclosporin group and Sappan group(P>0.05).Conclusion:The ethyl acetate extract of Sappan can improve cardiac function of mice with chronic viral myocarditis and inhibit myocardial hypertrophy.The ethyl acetate extract of Sappan can inhibit the expression of p38MAPK mRNA in myocardium of mice with chronic viral myocarditis,which may be one of the mechanisms of its treatment of chronic viral
