摘　　要：目的了解不同提取形式和不同核酸提取试剂盒对新型冠状病毒核酸提取效果的影响,为新型冠状病毒检测提供技术参考。方法采集2020年2月重庆市涪陵区疾控中心新型冠状病毒检测样本90份,均进行核酸提取。分析江苏硕世(机提1)和上海之江(机提2)、QIAGEN RNeasy Mini Kit RNA(手提1)和威斯腾(手提2)的提取效果。结果在阳性样本的检出率方面,机提1检出率为100.00%(10/10)和机提2检出率为90.00%(9/10)(P>0.05),手提1和手提2检出率均为100.00%(10/10)(P>0.05),机提和手提差异无统计学意义(P>0.05)。将手提1提取视为参考标准,在提取RNA总浓度和Ct值方面,手提2与之相比差异无统计学意义(P>0.05),而机提Ct值与之相比更大,差异有统计学意义(P<0.01)。结论新型冠状病毒核酸提取中,江苏硕世和上海之江、QIAGEN RNeasy Mini Kit RNA和威斯腾均有良好的提取效果,但在Ct值方面存在一定的差异,手提相对机提效果更好。Objective To understand the effects of different extraction forms and different nucleic acid extraction kits on the nucleic acid extraction of novel coronavirus,so as to provide technical reference for the detection of novel coronavirus.Methods A total of 90 samples of novel coronavirus from Fuling district center for disease control and prevention in February 2020 were collected for nucleic acid extraction.The extraction effects of Jiangsu Bioperfectus(mechanical extract 1)and Shanghai Liferiver(mechanical extract 2),QIAGEN RNeasy Mini Kit RNA(Handheld 1)and Wisten(Handheld 2)were analyzed.Results In terms of the detection rate of positive samples,it was 100.00%(10/10)of machine 1 and 90%(9/10)of machine 2(P>0.05).The detection rate of handheld 1 and handheld 2 were both 100.00%(10/10)(P>0.05).There was no statistically significant difference between mechanical extract and handheld(P>0.05).The extraction of handheld 1 was regarded as the reference standard,compared with the handheld 2 and handheld 1,there was no statistically significant difference in the total concentration of extracted RNA and the Ct value(P>0.05),while the mechanical extract Ct value was larger than that,and the difference was statistically significant(P<0.01).Conclusion In the extraction of novel coronavirus nucleic acid,Jiangsu Bioperfectus and Shanghai Liferiver,QIAGEN RNeasy Mini Kit RNA(Handheld 1)and Wisten all have good extraction results,but there are some differences in the Ct values.And the effect of handheld is better than machine lifted extraction.
