摘 要:目的探讨川芎嗪对长期注射水杨酸钠致听损伤的耳毒性拮抗作用。方法 50只健康级白色雄性SD大鼠随机分为生理盐水对照组(SC组,16只,每天腹腔注射生理盐水2.5 ml/kg)、水杨酸钠组(SS组,17只,每天腹腔注射400 mg/kg水杨酸钠溶液)和川芎嗪治疗组(TMP+SS组,17只,每天先腹腔注射400 mg/kg水杨酸钠溶液后,立即在对侧腹腔注射40 mg/kg盐酸川芎嗪溶液),各组连续注射14天后,在第15天比较各组体重、畸变产物耳声发射(DPOAE)幅值、听性脑干反应(ABR)阈值和潜伏期。结果第15天,SS组、TMP+SS组与SC组相比,体重明显减轻,差异有统计学意义(P<0.05);在f_2为1 000、2 000、3 000、4 000、6 000 Hz时,SS组DPOAE幅值较SC组升高(P<0.05);TMP+SS组与SC组相比,DPOAE幅值差异无统计学意义(P>0.05);在f_2为1 000、4 000、6 000 Hz时,SS组DPOAE幅值较TMP+SS组升高(P<0.05);SS组与SC组相比,ABR阈值升高,波Ⅱ潜伏期延长,差异有统计学意义(P<0.05),TMP+SS组与SC组比较,波Ⅱ潜伏期延长,差异有统计学意义(P<0.05)。结论川芎嗪对长期注射水杨酸钠致听损伤耳毒性的拮抗作用较弱,能部分拮抗长期注射水杨酸钠引起的DPOAE幅值增高,但对水杨酸钠导致的ABR阈值和潜伏期变化的拮抗作用还需要进一步研究。Objective To investigate the antagonistic effect of tetramethylpyrazine on the ototoxicity of long-term injection of sodium salicylate.Methods A total of 50 healthy white male SD rats were randomly divided into saline control group(SC group,2.5 ml/kg saline was injected intraperitoneally per day,16 rats),sodium salicylate group(SS group,400mg/kg sodium salicylate solution per day,17 rats)and tetramethylpyrazine treatment group(TMP+SS group,400 mg/kg sodium salicylate solution,with 40 mg/kg tetramethylpyrazine hydrochloride solution injected into the opposite abdominal cavity per day,17 rats).All groups were injected continuously for 14 days.On the 15th day,body weight,DPOAE amplitude,ABR threshold and latency were compared.Results①On the 15th day,compared with the saline control group,the weight of SS group and TMP+SS group decreased significantly,the difference was statistically significant(P<0.05).②On the 15th day,when f 2 was 1000 Hz,2000 Hz,3000 Hz,4000 Hz and 6000 Hz,DPOAE amplitude of SS group was higher than that of saline control group(P<0.05).There was no significant difference in DPOAE amplitude between TMP+SS group and saline control group(P>0.05).Compared with TMP+SS group,DPOAE amplitude of SS group increased at 1000 Hz,4000 Hz and 6000 Hz(P<0.05).③On the 15th day,compared with SC group,SS group had higher ABR threshold and prolongedⅡwave latency,the difference was statistically significant(P<0.05).Compared with SC group,TMP+SS group had longerⅡwave latency,and the difference was statistically significant(P<0.05).Conclusion Tetramethylpyrazine has a weak antagonistic effect on long-term injection of sodium salicylate ototoxicity.Tetramethylpyrazine can partially antagonize the increase of DPOAE amplitude caused by long-term injection of sodium salicylate,but the antagonism of tetramethylpyrazine to ABR threshold and latency needs further study.
