摘  要:目的探讨CUE结构域蛋白2(CUEDC2)对高糖诱导的肾小管上皮细胞损伤的影响。方法肾小管上皮细胞HK-2分为高糖处理组(HG)、转染对照载体后给予高糖处理组(NC+HG)、转染过表达CUEDC2载体后给予高糖处理组(CUEDC2+HG)及对照组(Con)。流式细胞仪测定细胞凋亡;试剂盒检测细胞中丙二醛(MDA)、超氧化物歧化酶(SOD)水平和培养液中TNF-α、IL-8水平。结果与Con组比较,HG组细胞凋亡率、MDA、TNF-α及IL-8升高[(6.23±0.65)%vs(18.56±3.14)%,(0.06±0.01)vs(0.15±0.02)nmol/mg,(29.54±3.56)vs(56.10±6.30)mg/L,(10.42±1.23)vs(19.59±1.68)mg/L;P<0.05],SOD活性降低[(36.20±2.78)vs(20.17±1.45)U/mg,P<0.05];与NC+HG、HG组比较,CUEDC2+HG组细胞凋亡率、MDA、TNF-α及IL-8降低[(17.14±2.98)%vs(18.56±3.14)%vs(11.47±1.20)%,(0.14±0.01)vs(0.15±0.02)vs(0.10±0.02)nmol/mg,(55.49±5.97)vs(56.10±6.30)vs(41.20±3.75)mg/L,(19.59±1.68)vs(20.12±2.35)vs(14.65±1.42)mg/L,P<0.05],SOD活性升高[(20.17±1.45)vs(19.64±1.37)vs(25.96±3.26)U/mg,P<0.05]。结论过表达CUEDC2可减少高糖诱导的肾小管上皮细胞炎症因子释放,降低细胞氧化应激,减少细胞凋亡,保护高糖诱导损伤的肾小管上皮细胞。Objective To investigate the effect of CUE domain protein 2(CUEDC2)on high glu-Methods HK-2 cells were divided into four groups:Con group,HG group,NC+HG group and CUEDC2+HG group.Apoptosis was detected by flow cytome-try.The levels of malondialdehyde(MDA),superoxide dismutase(SOD)of cells,TNF-αand IL-8 levels The apoptosis rate[(6.23±0.65)%vs(18.56±3.14)%]was higher;MDA content[(0.06±0.01)vs(0.15±0.02)nmol/mg]increased;TNF-α[(29.54±3.5)vs(56.10±6.30)mg/L]and IL-8[(10.42±1.23)vs(19.59±1.68)mg/L]increased;while SOD activity[(36.20±2.78)vs(20.17±1.45)U/mg]decreased in HG group than in Con group(P<0.05).The apoptotic rate[(11.47±1.20)%vs(17.14±2.98)%vs(18.56±3.14)%]was lower;MDA content[(0.10)±0.02]vs(0.14±0.01)vs(0.15±0.02)nmol/mg]decreased;TNF-α[(41.20±3.75)vs(55.49±5.97)vs(56.10±6.30)mg/L]and IL-8[(14.65±1.42)vs(20.12±2.35)vs(19.59±1.68)mg/L decreased,while SOD activity[(25.96±3.26)vs(19.64±1.37)vs(20.17±1.45)U/mg]increased in CUEDC2+HG group than in NC+HG and HG group(P<0.05).ConclusionOver-expression of CUEDC2 lowered high glucose-induced inflammatory factor released by renal tubular epithelial cells,decreased cellular oxidative stress,reduced apoptosis,and protected against high glucose-induced renal tubular epithelial cell injury.
