摘 要:目的通过对不同布鲁氏菌疫苗免疫接种途径与免疫效果之间的关系来评价商品化布鲁氏菌疫苗免疫羊后非应答组与应答组之间细胞因子分泌水平的影响,进一步探索其合理的免疫方案。方法采用试管凝集试验(SAT)和虎红平板凝集试验(RBPT)方法检测羔羊进行疫苗免疫后是否产生抗体分为非应答组和应答组,并采用酶联免疫吸附试验(ELISA)方法检测羊血清中IL-2、IL-4、IL-6、IL-10、MCP-1和IFN-γ细胞免疫水平。结果羔羊灌服接种S2疫苗免疫14 d内,非应答组细胞因子IL-2、IL-4、IL-6、IL-10、MCP-1和IFN-γ表达量均高于对照组,且IL-10表达量极显著升高(P<0.01),在免疫14 d时MCP-1的表达量显著升高(P<0.05);与非应答组相比,应答组IL-2、IL-4、IL-6和IL-10表达量均降低,MCP-1表达量升高,IFN-γ表达量则差异不大,且IL-2表达量极显著降低(P<0.01),在免疫7 d时IL-4和IL-6的表达量显著降低(P<0.05)。灌服接种M5疫苗免疫14 d内,非应答组与对照组相比,细胞因子IL-2、IL-4、IL-6和MCP-1的表达量先降低后升高,IL-10和IFN-γ表达量高于对照组;与非应答组相比,应答组IL-6和IFN-γ表达量升高,IL-4、IL-10和MCP-1先升高后降低,IL-2表达量则极显著降低(P<0.01),IL-4、IL-10、在免疫7 d时MCP-1和IFN-γ的表达量极显著升高(P<0.01)。皮下接种M5疫苗免疫14 d内,非应答组细胞因子IL-10、MCP-1和IFN-γ表达量均高于对照组,IL-2和IL-4,IL-6表达量先降低后升高,IFN-γ和MCP-1表达量显著升高(P<0.05);与非应答组相比,应答组IL-2、IL-6和IL-10的表达量均降低,IL-4的表达量升高,MCP-1和IFN-γ表达量则差异不大,且IL-2表达量极显著降低(P<0.01),在免疫14 d时IL-4的表达量显著升高(P<0.05)。皮下接种A19疫苗免疫14 d内,非应答组IL-4、IL-6、IL-10、MCP-1表达量均低于对照组,IL-2和IFN-γ表达量先升高后降低;与非应答组相比,应答组IL-4、IL-10、MCP-1和IFN-γ表达量均升高,IL-6表达量先�To evaluate the effect of different commercial Brucella vaccines on serum cytokines secretion in sheep of on-responding group and responding group, and to further explore a reasonable immune program.Sheep were divided into non-responding group and responding group according the results of tube agglutination test(SAT)and rose bengal plate test(RBPT) detecting the lambs serum antibodies after vaccination.The levels of IL-2,IL-4, IL-6, IL-10, MCP-1 and IFN-γ in sheep serum were detected by enzyme-linked immunosorbent assay(ELISA).Data showed that within 14 days of oral immunization with lamb S2 vaccine, the expression of IL-2, IL-4, IL-6, IL-10, MCP-1 and IFN-γ in the non-responding group were significantly higher than those in the control groups;the expression of IL-10 was significantly increased(P<0.01), while the expression of MCP-1 was significantly increased on the 14 th day of immunization(P<0.05);compared with the non-responding group, the expression levels of IL-2, IL-4, IL-6 and IL-10 were decreased in the responding group, while the expression of MCP-1 was increased, the expression of IFN-γ was not significantly changed, the expression of IL-2 was extremely decreased(P<0.01), and the expression levels of IL-4 and IL-6 were significantly different at 7 d(P<0.05).Within14 days of oral immunization with M5 vaccine, the expression of IL-2, IL-4, IL-6 and MCP-1 decreased first and then increased in the non-responding group compared with the control groups, while the expression of IL-10 and IFN-γ were higher than those in the control group;compared with the non-responding group, the expression levels of IL-6 and IFN-γ were increased, while IL-4, IL-10 and MCP-1 were increased first and then decreased, the expression of IL-2 was decreased extremely(P<0.01), and the expression levels of IL-4, IL-10, MCP-1 and IFN-γwere significantly different at 7 d immunization(P<0.01).Within 14 days after subcutaneous vaccination with M5 vaccine, the expression of IL-10, MCP-1 and IFN-γ in non-responding group were higher
