摘 要:目的:观察增液润燥汤对干燥综合征(Sjogren′s syndrome,SS)模型小鼠颌下腺干扰素-γ(interferon-γ,IFN-γ)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)及水通道蛋白5(aquaporin 5,AQP5)的影响,探讨该方治疗干燥综合征的作用机制。方法:将50只BALb/c小鼠,随机均分为5组:分别为模型对照组,增液润燥汤低剂量组、中剂量组、高剂量组,白芍总苷组,同时选取10只BALb/c小鼠作为正常组。免疫诱导法制备SS模型;酶联免疫吸附法检测颌下腺IFN-γ、TNF-α,免疫组织化学法、蛋白免疫印迹法检测AQP5的表达。结果:(1)与正常组比较,模型对照组、增液润燥汤低剂量组、白芍总苷组颌下腺IFN-γ、TNF-α水平升高(P<0.05);增液润燥汤中剂量组、高剂量组颌下腺IFN-γ、TNF-α水平升高,差异无统计学意义(P>0.05)。与模型对照组比较,增液润燥汤各剂量组与白芍总苷组颌下腺IFN-γ、TNF-α水平降低,差异均有统计学意义(P<0.05)。与白芍总苷组比较,增液润燥汤高剂量组颌下腺TNF-α水平降低,差异有统计学意义(P<0.05)。(2)与正常组比较,模型对照组AQP5表达明显减少(P<0.05);与模型对照组比较,增液润燥汤各剂量组、白芍组AQP5表达均增加(P<0.05),其中高剂量组AQP5表达接近正常组。结论:增液润燥汤可降低SS模型小鼠血清中IFN-γ、TNF-α的水平,升高AQP5的水平,可间接抑制Th1细胞的分化与增殖。Objective:To observe the effect of Zengye Runzao Decoction on IFN-γ,TNF-αand aquaporin-5 in submandibular gland of Sjogren′s syndrome(SS)model mice,and to explore the mechanism of its treatment.Methods:50 BALb/c mice were randomly divided into five groups:model control group,Zengye Runzao Decoction low-dose group,Zengye Runzao Decoction medium dose group,Zengye Runzao Decoction high-dose group,total glucosides of paeony group.Meanwhile,10 BALB/c mice were selected as normal group.SS model was established by immunoinduction;interferon-γ(IFN-γ)and tumor necrosis factor-α(TNF-α)in submandibular gland were detected by enzyme-linked immunosorbent assay,and the expression of aquaporin 5(AQP5)was detected by immunohistochemistry and Western blot.Results:(1)Compared with the normal group,the levels of IFN-γand TNF-αin submandibular gland of model control group,Zengye Runzao Decoction low-dose group and total glucosides of paeony group were increased(P<0.05);the levels of IFN-γand TNF-αin submandibular gland of middle dose group and high-dose group of Zengye Runzao Decoction were increased,and the difference was not statistically significant(P>0.05).Compared with the model control group,the levels of IFN-γand TNF-αin submandibular gland of each dose group of Zengye Runzao Decoction and total glucosides of paeony group were decreased,and the differences were statistically significant(P<0.05).Compared with the total glucosides of paeony group,the level of TNF-αin submandibular gland of Zengye Runzao Decoction high-dose group was significantly lower(P<0.05).(2)Compared with the normal group,the expression of AQP5 in the model control group was significantly decreased(P<0.05);compared with the model control group,the expression of AQP5 in each dose group of Zengye Runzao Decoction and the total glucosides of paeony group was increased(P<0.05),and the expression of AQP5 in the high-dose group was close to that in the normal group.Conclusion:Zengye Runzao Decoction can reduce the levels of IFN-γand TNF-αin seru
