摘  要:目的:观察鱼藤素通过调控Slit2/Robo2/Srgap2/Rac1信号通路对肺腺癌A549细胞增殖、侵袭和迁移能力的影响。方法:实验分为对照组(正常培养A549细胞)、鱼藤素低剂量组(A549细胞+10μmol·L-1鱼藤素)、鱼藤素中剂量组(A549细胞+30μmol·L-1鱼藤素)及鱼藤素高剂量组(A549细胞+50μmol·L-1鱼藤素)。通过噻唑蓝比色法检测细胞增殖抑制率;Transwell检测细胞侵袭能力;划痕实验检测细胞迁移能力;纤维状肌动蛋白(filamentous actin,F-actin)染色检测细胞骨架蛋白F-actin伪足面积;蛋白免疫印迹法检测Slit2、Robo2、Srgap2、Rac1信号通路蛋白及基质金属蛋白酶(matrix metalloproteinase,MMP)-2、MMP-9蛋白表达。结果:与对照组比较,鱼藤素低剂量组、中剂量组、高剂量组细胞增殖抑制率升高,细胞穿膜个数和划痕闭合情况降低,细胞F-actin伪足面积减少,Slit2、Robo2、Srgap2、Rac1、MMP-2及MMP-9蛋白表达降低,差异均有统计学意义(P<0.05)。结论:鱼藤素可能通过减少MMP-2、MMP-9蛋白表达影响细胞骨架蛋白F-actin数量和形态,降低肺腺癌A549细胞增殖、侵袭和迁移能力,其机制可能与下调Slit2/Robo2/Srgap2/Rac1信号通路相关。Objective:To observe the effect of deguelin on the proliferation,invasion and migration of lung adenocarcinoma A549 cells by regulating the Slit2/Robo2/Srgap2/Rac1 signaling pathway.Methods:The experiment was divided into control group(normally cultured A549 cells),low-dose deguelin group(A549 cells+10μmol·L-1 detolin),medium-dose deguelin group(A549 cells+30μmol·L-1 Detolin)and high-dose deguelin group(A549 cells+50μmol·L-1 detolin).Detection of cell proliferation inhibition rate by thiazole blue colorimetry,transwell detection of cell invasion ability,scratch test detection of cell migration ability,filamentous actin(F-actin)staining detection of cytoskeletal protein F-actin pseudopod area,Western blot detection of Slit2,Robo2,Srgap2,Rac1 signaling pathway proteins and matrix metalloproteinase(MMP)-2,MMP-9 protein expression were carried out.Results:Compared with that of the control group,the inhibition rate of cell proliferation increased in the low-dose,middle-dose and high-dose groups of deguelin,the number of cells penetrating the membrane and the closure of scratches were reduced,the area of cell F-actin pseudopods was reduced,Slit2,Robo2,Srgap2,Rac1,MMP-2 and MMP-9 protein expression decreased,the difference was statistically significant(P<0.05).Conclusion:Deguelin may affect the number and morphology of cytoskeleton protein F-actin by reducing the expression of MMP-2 and MMP-9 proteins,and reduce the proliferation,invasion and migration of lung adenocarcinoma A549 cells.The mechanism may be related to the down-regulation of Slit2/Robo2/Srgap2/Rac1 signaling pathway is related.
