摘　　要：目的:建立HPLC梯度洗脱法同时测定舒心降脂片中羟基红花黄色素A、红花黄色素A、3'-羟基葛根素、葛根素、3'-甲氧基葛根素、隐丹参酮、丹参酮Ⅰ和丹参酮ⅡA的含量。方法:采用Kromasil C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸溶液,梯度洗脱,流速1.0 ml·min^-1,柱温30℃,检测波长分别为:403 nm(检测羟基红花黄色素A和红花黄色素A)、250 nm(检测3'-羟基葛根素、葛根素和3'-甲氧基葛根素)和280 nm(检测隐丹参酮、丹参酮Ⅰ和丹参酮ⅡA)。结果:羟基红花黄色素A、红花黄色素A、3'-羟基葛根素、葛根素、3'-甲氧基葛根素、隐丹参酮、丹参酮Ⅰ和丹参酮ⅡA分别在4.17~83.40μg·ml^-1,2.86~57.20μg·ml^-1,2.49~49.80μg·ml^-1,8.93~178.60μg·ml^-1,2.26~45.20μg·ml^-1,0.69~13.80μg·ml^-1,4.46~89.20μg·ml^-1和3.67~73.40μg·ml^-1范围内线性关系良好(r≥0.9991),平均加样回收率分别为98.86%,99.01%,98.05%,100.05%,97.73%,96.97%,99.76%和98.35%,RSD分别为1.29%,1.13%,0.87%,0.97%,1.31%,1.26%,0.80%和1.44%(n=9)。结论:该方法操作简便、重复性好,可用于评价舒心降脂片的质量控制。Objective:To establish an HPLC gradient elution method for the simultaneous content determination of hydroxysafflor yellow A,safflor yellow A,3'-hydroxy puerarin,puerarin,3'-methoxy puerarin,cryptotanshinone,tanshinoneⅠand tanshinoneⅡA in Shuxin Jiangzhi tablets.Methods:The determination was performed on a Kromasil C18 chromatographic column(250 mm×4.6 mm,5μm)with the mobile phase consisting of acetonitrile-0.1%phosphoric acid solution with gradient elution at the flow rate of 1.0 ml·min-1.The detection wavelengths were set at 403 nm for hydroxysafflor yellow A and safflor yellow A,250 nm for 3'-hydroxy puerarin,puerarin and 3'-methoxy puerarin,and 280 nm for cryptotanshinone,tanshinoneⅠand tanshinoneⅡA.The column temperature was set at 30℃.Results:Hydroxysafflor yellow A,safflor yellow A,3'-hydroxy puerarin,puerarin,3'-methoxy puerarin,cryptotanshinone,tanshinoneⅠand tanshinoneⅡA showed a good linear relationship within the range of 4.17-83.40μg·ml^-1,2.86-57.20μg·ml^-1,2.49-49.80μg·ml^-1,8.93-178.60μg·ml^-1μg·ml^-1,2.26-45.20μg·ml^-1,0.69-13.80μg·ml^-1,4.46-89.20μg·ml^-1 and 3.67-73.40μg·ml^-1(r≥0.9991),respectively,and the average recovery was 98.86%,99.01%,98.05%,100.05%,97.73%,96.97%,99.76%and 98.35%with the RSDs of 1.29%,1.13%,0.87%,0.97%,1.31%,1.26%,0.80%and 1.44%(n=9),respectively.Conclusion:The method is easy to operate and repeatable,which can be used as a quality control method for Shuxin Jiangzhi tablets.
