摘 要:目的以铜绿假单胞菌为例,利用16S rDNA测序技术进行细菌同源基因分析应用价值探讨,为监测院内感染提供实验室依据。方法收集首都医科大学附属北京地坛医院分离的65株铜绿假单胞菌,采用16S测序技术,PCR(polymerase chain reaction)扩增16S rDNA 27-1492基因片段,测序结果利用MEGA-X软件绘制65株铜绿假单胞菌16S rDNA基因之间的进化关系图来表示。结果65株铜绿假单胞菌的亲缘分析中,菌株之间均有同源性,其中来自ICU的菌株,与本科室、神经外科、消化内科和呼吸科的菌株具有更近的亲缘性;呼吸科内也在不同患者标本中发现亲缘较近的菌株,其余科室均有与ICU和呼吸科同源的菌株。结论可应用16S测序技术对铜绿假单胞菌同源相关性进行监测,监测院内感染的发生。Objective The application value of 16S rDNA sequencing technology in analyzing homologous genotype of Pseudomonas aeruginosa in a hospital was evaluated to provide a laboratory basis for the hospital infection control.Methods The Pseudomonas aeruginosa isolated from Beijing Ditan Hospital Capital Medical University were collected and identified by 16S rDNA sequencing technology and PCR(Polymerase Chain Reaction).The sequences of 27F and 1492R were analyzed by using the MEGA-X software.Results 65 Pseudomonas aeruginosa strains were identified,in which the strains from ICU were more closely related to those from the department of neurology,neurosurgery,gastroenterology and respiratory medicine.In the laboratory of the department of respiration,closely related strains were also identified in the specimens of different patients.The other departments had strains of the same origin with ICU and the department of respiration.Conclusion The technology of 16S sequencing can be used to monitor the homologous correlation of Pseudomonas aeruginosa to prevent the outbreak of nosocomial infection.
