七味白术散总苷的提取工艺优化及体外抑菌活性初探

摘　　要：目的比较优化七味白术散总苷的提取工艺及初步探究其体外抑菌活性。方法采用均匀设计法,以乙醇浓度、乙醇用量、提取时间、超声功率为考察因素,以总苷量及人参皂苷量加权得到的综合评分为评价指标,通过二次逐步回归分析筛选水提醇沉法及超声辅助提取法从七味白术散汤剂中提取总苷的最优提取工艺,并对七味白术散总苷在体外抑制大肠杆菌及酵母菌的活性进行初步研究。结果研究得到水提醇沉法的优化提取工艺为:用14倍量的68%乙醇提取15 h,该工艺的浸膏得率为29.28%,并得到总苷0.28 g,人参皂苷0.0871 g。超声辅助提取法的优化提取工艺为:4倍量的56%乙醇在60%超声功率下提取40 min,浸膏得率为31.99%,得到总苷0.30 g,人参皂苷0.0917 g。结论综合考虑成本及提取效率,选择超声辅助提取法的最优工艺作为七味白术散总苷的制备方法。七味白术散总苷对大肠杆菌(Escherichia coli)及酵母菌(Saccharomycetes)均表现出抑制作用。Objective To optimize the optimal extraction conditions for total glycosides of Qiwei Baizhu powder.Methods Alcohol sedimentation and ultrasonic-assisted extraction were used.The extracting process was optimized by U6*(64)uniform design.Ethanol concentration,ethanol consumption,extractive times and ultrasonic power were investigated as important factors.Extract rate,total glycosides and ginsenoside were considered as the evaluation indices.The antibacterial effect in vitro of total glycosides in Qiwei Baizhu powder was also considered.Results The optimization process were predicted by DPS 13.5 based on quadratic stepwise regression analysis.The optimization process of total glycosides in Qiwei Baizhu powder which extracted by alcohol sedimentation was 14 times of 68%ethanol solution for 15 h.Under the conditions,the extract rate,total glycosides and ginsenoside was 29.28%,0.28 g and 0.0871 g,respectively.The optimization process of ultrasonic-assisted extraction was 4 times of 56%ethanol solution for 40 min under 60%ultrasonic power.Under the conditions,the extract rate,total glycosides and ginsenoside was 31.99%,0.30 g and 0.0917 g,respectively.Conclusion Considering the cost and extract efficiency,the optimal ultrasonic-assisted extraction was the best extraction method of total glycosides in Qiwei Baizhu powder.The total glycosides in Qiwei Baizhu powder showed inhibitory effects on Escherichia coli and Saccharomycetes.
