摘  要:目的探讨姜黄素去卵巢大鼠骨质疏松的作用及其对FoxO3a/Wnt信号通路的影响。方法于我院2018年3月~2019年5月通过大鼠双侧卵巢摘除复制绝经后大鼠骨质疏松动物模型开展研究,实验分为对照组、模型1组、白藜芦醇组[体重为5mg/(kg·d)]、姜黄素组[体重为20mg/(kg·d)],均灌胃给药,每日1次,连续8周后,采用ELISA法检测大鼠血清CT、OPG、BGP、RANKL、MDA、CAT、SOD含量;荧光探针DCFH-DA检测大鼠骨髓组织ROS水平;双能X射线骨密度检测仪分别测定股骨和腰椎BMD;三点弯曲法测量胫骨生物力学指标;Westernblot检测大鼠股骨组织中FoxO3a、β-catenin蛋白表达;qRT-PCR检测大鼠股骨组织中Gadd45α、Axin2基因表达。结果与模型1组比较,姜黄素组CT、OPG、SOD、CAT水平显著升高,但BGP、RANKL、MDA、ROS水平显著降低,差异有统计学意义(P<0.05);姜黄素升高腰椎/股骨BMD、胫骨最大载荷及最大应力,差异有统计学意义(P<0.05);姜黄素组FoxO3a蛋白及Gadd45α基因表达水平降低,β-catenin蛋白和Axin2基因的表达水平上升,与模型1组比较,差异有统计学意义(P<0.05)。结论姜黄素可通过激活FoxO3a/Wnt信号通路,有效缓解氧化应激介导的去卵巢大鼠骨质疏松。Objective To investigate the inhibiting function of curcumin on osteoporosis in ovariectomized rats and its impact on FoxO3a/Wnt signaling pathway.Methods From March 2018 to May 2019,an animal model of postmenopausal osteoporosis was established by bilateral ovariectomy in rats.They were divided into the control group,the model group 1,the resveratrol group(body weight was 5mg/[kg·d])and the curcumin group(body weight was 20mg/[kg·d]).All rats were given intragastric administration,qd.After 8 weeks,the serum contents of CT,OPG,BGP,RANKL,MDA,CAT and SOD of rats were detected by ELISA.ROS level in rat bone marrow tissue was detected by fluorescence probe DCFH-DA.Dual-energy X-ray bone mineral density detector was used to measure BMD of femur and lumbar vertebrae respectively.Biomechanical indexes of tibia were measured by three-point bending method.Westernblot was used to detect the expression of FoxO3a and β-catenin proteins in rat femoral tissue.qRTPCR was used to detect the expression of Gadd45α and Axin2 genes in rat femoral tissue.Results Compared with the model group 1,the levels of CT,OPG,SOD and CAT in the curcumin group significantly increased,while the levels of BGP,RANKL,MDA and ROS significantly decreased,with statistically significant differences(P<0.05).Curcumin increased the BMD of lumbar spine/femur,maximum load and maximum stress of tibia,and the differences were statistically significant(P<0.05).In the curcumin group,the expression level of FoxO3a protein and Gadd45αgene increased,while the expression level ofβ-catenin protein and Axin2 gene decreased,compared with the modelgroup 1 with statistically significant differences(P<0.05).Conclusion Curcumin can effectively alleviate oxidative stress-mediated osteoporosis in ovariectomized rats by activating FoxO3a/Wnt signaling pathway.
