摘 要:目的探讨影响山药中薯蓣皂素得率的重要因素,旨在为山药中薯蓣皂素的综合利用,特别是提出过程研究提供科学依据。方法利用闪氏提取法提取山药中的薯蓣皂素,在提取溶剂单因素实验的基础上,选择料液比、提取时间和次数进行正交实验,并进行优化和验证。结果采用2次闪式提取法对山药中的薯蓣皂素进行提取,最佳工艺条件为:5%硫酸的80%乙醇溶液,料液比1∶10,闪式提取1次,提取时间2min,加热水解2h,离心,取上清液,回收溶剂,收集残渣,冷却,使用1%碳酸钠溶液中和,离心得到滤渣,将滤渣置于干燥皿在80℃烘箱中干燥,所得水解物以石油醚进行提取,此时的料液比为1∶10,再次闪式提取1次,提取时间2min。结论在此条件下,薯蓣皂素得率达到8.14mg/g。闪氏提取法简单、迅速、高效,可用于山药中薯蓣皂素的提取。Objective To discuss the important factors that affect the yield of diosgenin in common yam rhizome so as to provide scientific basis for the comprehensive utilization of diosgenin in common yam rhizome,especially for the process research.Methods The diosgenin in commonyam rhizome was extracted by using the flash extraction method.Based on the single factor experiment of the extraction solvent,the ratio of material to liquid,extraction time and frequency were selected for orthogonal experiment,and optimization and validation were carried out.Results The diosgenin in commonyam rhizome was extracted by two flash extraction methods.The best process conditions were:5%sulfuric acid in 80%ethanol solution,material-to-liquid ratio 1∶10.After flash extraction once for 2 min,heat and hydrolyze for 2 hours,centrifuge,take the supernatant,recover the solvent,collect the residue,cool,neutralize with 1%sodium carbonate solution,centrifuge to obtain the filter residue,place the filter residue in a drying dish and dry in an oven at 80℃,and the hydrolysate obtained was extracted with petroleum ether,the material-liquid ratio was 1∶10 at this time,flash extraction once again,extraction time 2min.Conclusion Under these conditions,the yield of diosgenin reached 8.14mg/g.The flash extraction method is simple,rapid,and highly effective,and can be used to extract diosgenin from commonyam rhizome.
