摘 要:目的:采用高通量测序技术鉴定星形胶质细胞外显子组中A至I RNA编辑。方法:对4个星形胶质细胞样本的转录组和外显子组分别进行高通量测序,利用生物信息学方法进行深度分析,鉴定出星形胶质细胞外显子组中A至I RNA编辑位点。结果:在星形胶质细胞外显子组中共鉴定出1751个A至I RNA编辑位点,其中5’UTR有110个,3’UTR有1390个,编码区有251个,包括118个错义A至I RNA编辑位点。结论:本研究完成了星形胶质细胞外显子组中A至I RNA编辑位点的鉴定,研究结果将有助于深入理解A至I RNA编辑在神经系统疾病发生中的作用机制。Objective:To analyze A-to-I RNA editing in exome of astrocyte by the high-throughput sequencing technique.Methods:Transcriptome and exome of four astrocyte samples were sequenced through high-throughput sequencing technology respectively.Bioinformatic analyses were applied for further study of the sequencing results to identificate A-to-I RNA editing sites in exome of astrocyte.Results:A total of 1751 A-to-I RNA editing sites in exome of astrocyte were identificated,of which 110 sites located on 5’UTRs,1390 sites located on 3’UTRs,251 sites located on CDS,including 118 missense mutations.Conclusion:This study completed identification of A-to-I RNA editing sites in exome of astrocyte.The research findings can contribute to the better understanding of the molecular mechanisms of A-to-I RNA editing in nervous system disease.
