摘 要:目的采用微透析取样技术研究口服给予天麻成分C后血-脑药代动力学参数。方法采用HPLC技术建立微透析样品中天麻成分C的分析方法,采用正透析法和反透析法研究天麻成分C在大鼠体内的血、脑微透析探针体内外回收率,考察灌流速度、药物质量浓度对探针体内外回收率的影响,以及探针回收率在大鼠体内外的稳定性。采用血-脑同步微透析进行体内药动学研究,HPLC测定透析液中天麻成分C的含量。结果所建立的HPLC分析方法,在要求范围内线性关系良好,色谱的专属性、精密度、基质效应等均符合要求。体内外回收率试验,采用正透析法和反透析法测得体外内回收率显示在恒定质量浓度下,血、脑微透析探针体内外回收率均随着流速的增大而减小,体外回收率基本保持一致,体内回收率低于体外回收率;恒定流速下,天麻成分C质量浓度对血脑探针体内外回收率均无影响;在恒定浓度和恒定流速的条件下,天麻成分C的血脑微透析探针在大鼠体内回收率在12 h内保持相对稳定,血脑探针回收率分别为(45.32±2.83)%,(10.55±0.82)%。结论反透析法可以用来研究天麻成分C在大鼠体内的回收率,微透析技术能够用于天麻成分C在大鼠体内的血、脑药代动力学同步研究。Objective:To study the blood-brain pharmacokinetic parameters after oral administration of Gastrodia elata component C by microdialysis sampling technique.Methods:HPLC was used to establish the analysis method of Gastrodia elata component C in microdialysis samples and the positive dialysis method and reverse dialysis method were used to study the recovery rate of Gastrodia elata component C in the blood-brain microdialysis probes in rats in vivo and in vitro,and to explore the effect of the perfusion speed and drug concentration on the recovery rate of the probe in vivo and in vitro and the stability of the recovery rate of the probe in vivo in rats.The pharmacokinetic study in vivo was carried out by blood-brain synchronous microdialysis and the content of Gastrodia elata component C in the dialysate was determined by HPLC.Results:The established HPLC analysis method had a good linear relationship within the required range and the specificity,precision and matrix effect of the chromatogram all meet the requirements.The recovery rate test in vivo and in vitro,by which the recovery rate was measured through positive dialysis method and reverse dialysis method,showed that the recovery rates in vivo and in vitro of blood-brain microdialysis probes all decreased with the increase of flow rate at a constant mass concentration.The recovery rate in vitro was basically the same,and the recovery rate in vivo was lower than the recovery rate in vitro.At a constant flow rate,the mass concentration of Gastrodia elata component C had no effect on the recovery rate of blood-brain probe in vivo and in vitro.Under the conditions of constant concentration and constant flow rate,the recovery rate of the blood-brain microdialysis probe of Gastrodiae elata component C in rats remained relatively stable within 12 hours.The recovery rate of the blood-brain probe was 45.32±2.83%and 10.55±0.82%,respectively.Conclusion:The reverse dialysis method can be used to study the recovery rate of Gastrodia elata component C in rats and the mic
