摘　　要：目的:研究TLR2胞内信号传导区域内Arg677Trp、Arg753Gln基因多态性是否与梅毒治疗后患者的血清学阳性结果相关。方法:采用PCR-RFLP对实验组1的100例梅毒血清固定患者、实验组2的50例梅毒治疗后TPPA阳性而TRUST阴性的患者及100例健康对照者进行检测;部分判定困难的标本进一步采用DNA测序证实。结果:所有样本均未检测到TLR2 Arg677Trp基因多态性。对照组无Arg753Gln基因多态性,实验组1有1例(1%)TLR2 Arg753Gln基因多态性,与对照组比较无明显差异(X^2=1.01,P=0.316);实验组2有6例(12%)TLR2 Arg753Gln基因多态性,与对照组比较有明显差异(X^2=12.50,P<0.01)。结论:目前未发现TLR2 Arg677Trp和Arg753Gln基因多态性与梅毒血清固定相关。梅毒规范治疗后出现TPPA阳性TRUST阴转的现象,可能与TLR2Arg753Gln基因多态性相关。Objective:To investigate whether the Arg677Trp and Arg753Gln polymorphisms in the intracellular signaling region of TLR2 are linked to post-treatment serological test positive in syphilis patients.Methods:PCR-RFLP was used to detect TLR2 Arg677Trp and Arg753Gln polymorphisms in 100 syphilis patients with sero-fixation(group 1),50 patients with TPPA positive while TRUST negative after syphilis treatment(group 2),and 100 healthy controls.DNA sequencing was used to confirm the diagnosis in case of diagnostic uncertainty.Results:TLR2 Arg677Trp polymorphism was not detected in any of the samples.While only one patient in group 1(1%)displayed TLR2 Arg753Gln polymorphism,proportion of patients with TLR2 Arg753Gln polymorphism was significantly higher in group 2 than in the controls(6/50 vs.0,x^2=12.50,P<0.01).Conclusions:Neither TLR2 Arg677Trp nor Arg753Gln polymorphisms is associated to syphilis serum fixation.However,TLR2 Arg753Gln polymorphism could be linked to post-treatment TPPA positive and TRUST negative in syphilis patients.
