摘　　要：目的研究蒲公英水煎液、蒲公英多糖及蒲公英萜醇3种蒲公英提取物对T47D、SK-BR-3乳腺癌细胞增殖作用的影响。方法 CCK-8检测法初步探究3种蒲公英提取物对SK-BR-3、T47D乳腺癌细胞增殖的抑制作用;实时xCELLigence细胞分析系统进行细胞增殖毒性检测;划痕愈合实验观察蒲公英萜醇对T47D细胞迁移能力的影响;流式细胞仪分析蒲公英萜醇对T47D乳腺癌细胞生长凋亡的影响。结果 3种蒲公英提取物对SK-BR-3、T47D乳腺癌细胞的增殖均有一定的抑制作用,对T47D乳腺癌细胞增殖的抑制作用更明显,其中蒲公英萜醇对2种乳腺癌细胞增殖的抑制作用大于蒲公英水煎液和蒲公英多糖;同时蒲公英萜醇能显著抑制T47D乳腺癌细胞增殖迁移并可诱导其凋亡,中位剂量下凋亡率可达16.17%~20.86%。结论蒲公英对乳腺癌细胞的增殖具有一定的抑制作用。Objective The effects of dandelion decoction, dandelion polysaccharide and dandelion sterol on the proliferation of T47D and SK-BR-3 breast cancer cells were studied. Methods CCK-8 assay for preliminary investigation of the inhibitory effects of three dandelion extracts on the proliferation of SK-BR-3 and T47D breast cancer cells;Real-time xCELLigence cell analysis system for cell proliferation toxicity testing;The effect of dandelion sterol on the migration ability of T47D cells was observed by scratch healing experiment;Flow cytometry analysis of the effects of dandelion sterol on the growth and apoptosis of T47D breast cancer cells. Results Three kinds of dandelion extracts have certain inhibitory effects on the proliferation of SK-BR-3 and T47D breast cancer cells,however, the inhibitory effect on the proliferation of T47D breast cancer cells is more obvious,Among them, dandelion sterol inhibited the proliferation of two breast cancer cells more than dandelion decoction and dandelion polysaccharide;At the same time, dandelion sterol can significantly inhibit the proliferation and migration of T47D breast cancer cells and induce apoptosis, Apoptosis rate can reach 16.17%-20.86% at median dose. Conclusions Dandelion has a certain inhibitory effect on the proliferation of breast cancer cells.
