摘  要:为研究口蹄疫兔化弱毒疫苗ZBatt株和其亲本强毒株ZBCF222之间的生物学特性差异,测定了ZBatt和ZBCF222病毒复制效率,病毒感染细胞后6种宿主因子mRNA表达水平,比较了病毒对乳鼠和豚鼠致病性差异。结果显示:ZBatt和ZBCF222分别感染PK、IBRS-2细胞均不产生明显CPE,感染BHK-21细胞均可以产生CPE和相似的蚀斑形态,病毒生长曲线无显著差异(P>0.05);ZBCF222感染牛原代肾细胞(BK)可产生CPE,形成明显蚀斑,接种豚鼠可产生致病性,而ZBatt感染BK细胞不产生CPE,对豚鼠无明显致病性;二者在BK细胞上的病毒生长曲线和RNA复制效率呈显著性差异(P<0.05);ZBCF222感染BK细胞后宿主因子表达水平与ZBatt感染差异均不显著(P>0.05)。本研究结果表明,亲本强毒株ZBCF222对天然宿主来源细胞具有强致病力,弱毒疫苗ZBatt则不具有致病力,豚鼠可作为试验动物替代牛用于ZB病毒毒力差异评价。The rabbit-attenuated ZBatt strain of foot-and-mouth disease(FMD)virus was obtained by serial 100 passages of the wild-type virulent ZB virus in suckling rabbits.In the present study,the ZBatt strain and its parental virulent ZBCF222 were compared for some biological properties and expression level of six host cell proteins.The results showed that the ZBatt and ZBCF222 were able to infect BHK-21 but not PK and IBRS-2 cells.The ZBatt strain showed similar plaque phenotypes and growth kinetics to ZBCF222 strain in BHK-21 cells.However,ZBCF222 strain was able to infect primary fetal bovine kidney(BK)cells and produced CPE with plaques while ZBatt strain did not.The viral growth kinetics and RNA replication levels of virulent ZBCF222 strain in BK cells were significantly higher than ZBatt strain(P<0.05).The mRNA relative expression levels of six host cell proteins CCL2,IFN-p,ISG15,Mxl,OAS and PKR in BK cells were no significant difierence(P>0.05)between the ZBatt and ZBCF222 strains.The survival analysis in infected suckling mice showed that the ZBatt strain exhibited higher virulence than ZBCF222 strain.However,ZBCF222 strain produced a remarkably pathogenical damages in the infected guinea pigs while ZBatt strain exhibited an attenuated phenotype.Our findings suggested that the biological properties of the rabbit attenuated ZBatt strain and its parental virulent strain were significantly different in natural host cells.Guinea pigs might be used to replace cattle as the laboratory animal model to evaluate virulence and pathogenicity of ZB viruses.
