摘 要:DNA易受各种内源性和外源性作用的干扰而出现损伤,这些损伤会引起遗传信息载体产生不可修复性的变化,从而导致癌症等重大疾病的发生。当DNA发生损伤时,大量的γ-H2AX会聚集于损伤位点,释放生物信号并引导修复蛋白对DNA进行修复,因此,γ-H2AX被认为是DNA损伤的重要标志物之一。本文结合国内外研究,对不同的DNA损伤类型进行了论述,同时对包括免疫荧光染色法、流式细胞术法、蛋白质印迹法和全细胞酶联免疫反应法在内的多种γ-H2AX检测方法进行了总结,为DNA损伤研究和γ-H2AX检测提供了借鉴与参考。DNA damage is susceptible resulted by the endogenous and exogenous effects,the damage is capable of causing irreparable changes in genetic information carriers,which further causes the occurrence of cancer and other serious diseases.Once the DNA is damaged,large amounts ofγ-H2 AX accumulated at the damage site to release biological information and guiding repair proteins to restore the DNA.Therefore,γ-H2AX is recognized as one of the important markers of DNA damage.Based on domestic and foreign literature,this review summarized different types of DNA damages;Meanwhile,the relevant determination methods including immunofluorescence,flow cytometry,western blot and whole cell enzyme-linked immunoreaction were discussed.This review provides beneficial support for the DNA damage study andγ-H2 AX determination.
