摘  要:目的建立人尿中1-羟基芘(1-OHP)荧光猝灭法的测定方法。方法在p H值为4.5的磷酸氢二钠-柠檬酸缓冲液中,1-OHP与鱼精蛋白-金纳米簇(PRT-Au NCs)反应,通过荧光猝灭程度(ΔF)得到标准曲线,对1-OHP进行定量。结果在最佳实验条件下,测定值为1.03 nmol/L^62.80 nmol/L时,1-OHP与ΔF有良好的线性关系,线性回归方程为ΔF=577.91+17.89 c(10-9mol/L),R2为0.998 5,方法检出限为0.309 nmol/L,人尿样中1-OHP的加标回收率为94.39%~104.52%。结论本法简便、快速、灵敏,所用仪器设备简单,易于推广,提高金纳米簇传感系统的灵敏度,可用于人尿中1-OHP的检测,为评价人体在多环芳烃(PAHs)下的暴露情况提供数据支持。Objective To establish a fluorescence quenching method for the determination of 1-hydroxypyrene( 1-OHP) in human urine.Methods 1-OHP reacted with protamine-gold nanocluster( PRT-Au NCs) in Na2 HPO4-citric acid buffer of p H 4.5.The standard curve was obtained by fluorescence quenching degree( ΔF),and the 1-OHP was quantified.Results Under the optimum experimental conditions,there was a good linearity between 1-OHP and ΔF when the determination was within 1.03 nmol/L-62.80 nmol/L.The linear regression equation was ΔF=577.91 + 17.89 c( 10-9 mol/L),R2 was 0.998 5.The detection limit of the method was 0.309 nmol/L,and the recoveries of added 1-OHP in the human urine samples were within 94.39%-104.52%.Conclusion The method is simple,rapid and sensitive,and easy to popularize.It can improve the sensitivity of gold nanocluster sensing system and can be used for the detection of 1-OHP in human urine.The study can provide data support for evaluating human exposure to polycyclic aromatic hydrocarbons( PAHs).
