摘 要:目的:探讨黄芪甲苷对氧糖剥夺/复氧复糖PC12细胞损伤的保护作用及机制。方法:选取对数生长期PC12细胞,随机分为正常对照组(Control)、模型组(Model)、黄芪甲苷组(AST-Ⅳ)。Control组细胞正常培养;Model组和AST-Ⅳ组细胞在氧糖剥夺2 h后,复氧复糖24 h;AST-Ⅳ组细胞在复氧复糖的同时给予黄芪甲苷(100 mmol/L)处理。倒置显微镜观察细胞生长状态,MTS法检测细胞存活率,Western blot检测Bax、Bcl-2和Caspase-3的表达。结果:与Control组相比,Model组细胞折光性差,突触减少,细胞肿胀聚集、脱落,存活率降低,Bcl-2表达减少,Bax、Caspase-3的表达增多(P<0.05)。与Model组比较,AST-Ⅳ组细胞生长状态好转,肿胀、聚集减少,存活率增高,Bax、Caspase-3的表达减少,Bcl-2表达增多(P<0.05)。结论:黄芪甲苷可通过抑制氧糖剥夺/复氧复糖PC12细胞的凋亡发挥保护作用。Objective:To investigate the protective effect and mechanism of AstragalosideⅣon PC12 cells injury after oxygen-glucose deprivation/reoxygenation.Methods:PC12 cells with logarithmic growth were randomly divided into the control group,the model group and AST-Ⅳgroup.The control group was cultured normally without any intervention.Cells in the model group and AST-Ⅳgroup were reoxygenated for 24 h after two hours of oxygen and sugar deprivation,and the cells in AST-Ⅳgroup were also treated with AstragalosideⅣ(100μmol/L)during reoxygenation.The growth status of the cells was observed by inverted microscope,the survival rate was detected by MTS method,and the expressions of Bax,Bcl-2 and Caspase 3 were detected by Western blot method.Results:The cells in the model group showed poor refractive index,decreased and disappeared synapses,cell swelling,aggregation and shedding,decreased cell survival rate,and there were increased expressions of Bax and Caspase3,as well as decreased expression of Bcl-2 in the model group compared to those in the control group(P<0.05).AST-Ⅳgroup showed improved growth of cells,reduced cell swelling and aggregation,increased cell survival rate,and decreased expressions of Bax and Caspase,as well as increased expression of Bcl-2 when compared to the model group(P<0.05).Conclusion:AstragalosideⅣplays a protective role by inhibiting the apoptosis of oxygen-glucose deprivation/reoxygenation PC12 cells.
