基于丁香苦苷抗菌药效研究的丁香叶质量标志物指认

摘　　要：目的:通过丁香苦苷对9种常见致病菌株(金黄色葡萄球菌、大肠杆菌、铜绿假单胞杆菌、白色葡萄球菌、肺炎杆菌、痢疾志贺氏菌、福氏志贺氏菌、宋内氏志贺氏菌、变形杆菌)的抑菌作用研究,并与5种常用抗菌阳性药(炎立消胶囊、炎可宁片、头孢克肟胶囊、黄连素片、双黄连口服液)抗菌效果对比,明确丁香苦苷的抗菌疗效,实现丁香叶质量标志物的指认。方法:采用倍比稀释法、打孔法、体外杀菌实验3种方法观察细菌生长状况,测定最低抑菌浓度(MIC)、最低杀菌浓度(MBC)和抑菌环直径。结果:倍比稀释法对选定的9个菌株均有抑菌作用,其中对白色葡萄球菌及肺炎杆菌抑菌作用强,MIC为3.125 mg·mL^(-1)及6.25 mg·mL^(-1 ),对大肠杆菌、铜绿假单胞杆菌抑制作用一般,MIC为50 mg·mL^(-1)。打孔法丁香苦苷对选定的9个菌种中的7个菌种出现抑菌环,但对大肠杆菌、铜绿假单胞杆菌没有抑菌环。体外杀菌实验结果表明对大部分菌株均有杀菌作用,其中对白色葡萄球菌和肺炎杆菌作用显著,MBC分别为3.125 mg·mL^(-1)和6.25 mg·mL^(-1),对痢疾志贺氏菌、福氏志贺氏菌、变形杆菌、金黄色葡萄球菌杀菌效果相似,对宋内氏志贺氏菌杀菌作用一般,在最大药物浓度下对大肠杆菌,铜绿假单胞杆菌没有杀菌作用。结论:丁香苦苷是一种广谱抑菌药物,与丁香叶功效相似,尤其是白色葡萄球菌、肺炎杆菌特别明显。Objective:To study the antimicrobial effects of syringopicroside on 9 common pathogenic strains(Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginosa,Staphylococcus albicans,Bacillus pneumoniae,dysentery Shigella,Shigella flexneri,Shigella sonnei,Proteusbacillus vulgaris),and to compare its antibacterial effects with the antibacterial effects of 5 kinds of commonly used antibacterial positive drugs(Yanlixiao Capsule,Yankening Tablet,Cefixime Capsule,Berberine Tablet,Shuanghuanglian Oral Liquid),thus to clarify the antibacterial effect of syringopicroside and to identify the quality markers of Folium syringgae.Methods:The growth status of bacteria was observed by using the method of double ratio dilution,punching method and in vitro sterilization experiment,and MIC,MBC and the diameter of bacteriostatic ring were determined.Results:The multiplication dilution method showed it had bacteriostatic effect on 9 selected strains,including the antimicrobial effect on Staphylococcus aureus and Bacillus pneumoniae,MIC was 3.125 mg·mL-1 and 6.25 mg·mL-1.It had the average inhibitory effect on Escherichia coli and Pseudomonas aeruginosa,and MIC was 50 mg·mL-1.There were bacteriostatic rings on 7 strains of 9 species selected,but there were no bacteriostatic rings for Escherichia coli and Pseudomonas aeruginosa.The results of in vitro sterilization showed that it had bactericidal effect on most strains,especially on Staphylococcus aureus and Bacillus pneumoniae,and MBC was 3.125 mg·mL-1 and 6.25 mg·mL-1 respectively;it had similar bactericidal effect on dysentery Shigella,Shigella flexneri,Proteusbacillus vulgaris and Staphylococcus aureus;it had average bactericidal effect on Songnei’s Shigella;it had no bactericidal effect on Escherichia coli and Pseudomonas aeruginosa at the maximum drug concentration.Conclusion:Syringopicroside is a kind of spectral bacteriostatic drug,its effect is similar to that of Folium syringgae.It has especially obvious effect on Staphylococcus aureus and Klebsiella pneumoniae.
