摘  要:目的建立高效液相色谱法测定雷公藤与南蛇藤中雷公藤红素含量的方法,并进行不同产地药材含量比较。方法采用95%乙醇热回流提取,色谱柱为Welch Ultimate XB-C18(5μm,4.6×250mm);流动相为乙腈(B)-0.1%甲酸,梯度洗脱,检测波长为425nm,柱温30℃,流速1.0mL·min^-1;进样量10μL。结果雷公藤红素浓度在0.124~49.75μg·mL^-1(r=0.9991)范围内线性关系良好,平均回收率为101.09%,RSD=1.89%,14批样品中,产于广西百色的南蛇藤中雷公藤红素含量最高的为1405.29μg·g^-1。结论建立的HP LC方法操作简便、稳定、准确且可行,可作为药材中雷公藤红素的定量分析方法,广西百色南蛇藤更适合作为雷公藤红素提取的植物来源。OBJECTIVE To establish a HPLC method for determination of celastrol in Tripterygium wilfordii Hook.f.and Celastrus orbiculatus Thunb.The cont ent of crude drugs from different producing areas was compared.METHODS Heat reflux extraction with 95% ethanol was used,Welch Ultimate XB-C 18(5μm,4.6×250mm)column was used,a mobile phase of acetonitrile(B)-0.1% formic acid solution,gradient elution,and the determined wavelength was set at 425nm,the column temperature was 30℃.The flow rate was 1.0mL·min^-1 and injection was 10μL.RESULTS Celastrol had a good linear relationship within 0.124-49.75μg·mL^-1(r=0.9991),the average recovery was 101.09% with the RSD 1.89%.Among the 14 samples,the highest content of celastrol in the Celastrus orbiculatus Thunb produced in Baise,Guangxi is 1405.29μg·g^-1.CONCLUSION The results indicated that the developed method would be simple,stable and accuracy and feasible.It can be used for quantitative analysis of celastrol in medicinal materials.Celastrus orbiculatus Thunb in Baise,Guangxi is more suitable as a plant source for celastrol extraction.
