摘 要:该试验旨研究在体外培养中添加甘草素(Liquiritigenin,LQ)对小鼠早期胚胎体外发育的影响,采集6周龄雌性昆明小鼠胚胎,随机分为对照组和不同浓度LQ处理组(0、10、20、30、40、50μmol/L)。试验通过囊胚率筛选出LQ最适浓度,然后检测小鼠胚胎氧化应激的3个指标,利用DCFH-DA检测早期胚胎内活性氧(reactive oxygen species,ROS)水平、利用CMF 2 HC检测早期胚胎内谷胱甘肽(glutathione,GSH)水平、采用JC-1检测早期胚胎线粒体膜电位水平。结果表明:20μmol/L的LQ处理组囊胚发育率显著高于0和10μmol/L的LQ处理组(P<0.05),且极显著高于30、40和50μmol/L的LQ处理组(P<0.01)。20μmol/L的LQ处理组ROS水平与对照组相比显著降低(P<0.05);20μmol/L LQ处理组GSH水平与对照组相比显著提高(P<0.05);20μmol/L LQ处理组线粒体膜电位水平与对照组相比显著提高(P<0.05)。结果表明,在体外培养过程中添加LQ可以提高小鼠胚胎发育率,降低胚胎内ROS水平,提高胚胎内GSH水平,改善早期胚胎线粒体功能,从而减少氧化应激带来的损伤并进一步提高小鼠早期胚胎发育的质量。The aim of this study is to study the effect of adding Liquiritigenin(LQ)to the in vitro development of mouse early embryos in vitro.6-week-old female Kunming mouse embryos were collected and randomly divided into control group and different concentrations of LQ treatment groups(0,10,20,30,40,50μmol/L).The experiment screened out the optimal concentration of LQ through the blastocyst rate,and then detected three indicators of oxidative stress in mouse embryos.DCFH-DA was used to detect the level of reactive oxygen species(ROS)in early embryos,and CMF2HC was used to detect early embryos.Glutathione(glutathione,GSH)level,JC-1 was used to detect early embryo mitochondrial membrane potential level.The results showed that the blastocyst development rate of the 20μmol/L LQ treatment group was significantly higher than that of the 0μmol/L and 10μmol/L LQ treatment groups(P<0.05),and was extremely significantly higher than that of the 30μmol/L,40μmol/L,and 50μmol/L LQ treatment groups(P<0.01).The ROS level of the 20μmol/L LQ treatment group was significantly lower than that of the control group(P<0.05);the GSH level of the 20μmol/L LQ treatment group was significantly higher than that of the control group(P<0.05);the mitochondrial membrane potential level of the 20μmol/L LQ treatment group was significantly improvedcompared with the control group(P<0.05).The results show that adding LQ during in vitro culture can increase mouse embryo development rate,reduce embryonic ROS level,increase embryonic GSH level,improve early embryonic mitochondrial function,thereby reducing damage caused by oxidative stress and further improving the quality of early embryonic development in mice.
