摘  要:目的探讨补肾活血软坚方对大鼠冠脉微血管病变内皮损伤相关因子的作用。方法将60只Wistar大鼠采用随机数字表法抽取15只作为假手术组,余作为造模组,造模组采用向大鼠左心室注射月桂酸钠的方法建立大鼠冠脉微血管内皮功能损伤动物模型,造模成功43只,随机分成4组,分别是:假手术组、模型组、喹那普利组、中药组,以假手术组为对照组。用药组分别给予喹那普利2 mg/(kg·d),中药方15 g/(kg·d)、假手术组和模型组给予等量的蒸馏水灌服,每日一次。选择用药后第28 d作为观察点,观察心肌病理切片并用ELISA检测血清中的NO、内皮素-1(ET-1)。结果与假手术组比较,模型组大鼠血清NO明显减少(P<0.01),ET-1明显升高(P<0.01);与模型组比较,中药组血清NO明显升高(P<0.01),ET-1明显减少(P<0.01);与喹那普利组比较,中药组血清NO明显升高(P<0.01),ET-1亦升高(P<0.05)。结论大鼠冠脉微血管病变内皮功能损伤,血清NO降低,ET-1增加;中药组可增加NO,降低ET-1,改善微血管病变内皮功能;用药28天,与喹那普利相比,中药组在增加NO及降低ET-1方面未见明显优势。Objective:To investigate the mechanism of Chinese herbal medicine with actions of invigorating kidney and activating blood and softening scleroma on related molecules of coronary microcirculation endothelial injury in rats with myocardial infarction.Methods:The 60 Wistar rats were randomly selected 15 as the sham operation group by random number table method,and others as modules.The rat model of coronary microcirculation endothelial injury with myocardial infarction was established by receiving apical left ventricular injection of sodium laurate.The 43 rats in the success of the manufacturing model were randomly divided into 4 groups:Sham-operation group,model group,quinapril group,Chinese herbal medicine group,and the Sham-operation team was the control group.Rats in the treatment groups were given Quinapril 2 mg/(kg·d),and Chinese herbal medicine decoction 15g/(kg·d);Rats in the control and model groups were given equivalent dose of distilled water one day.The pathological sections of myocardium were observed and the levels of serum Nitricoxide(NO)and endothelin-1(ET-1)were detected by using enzyme-linked immunosorbent assay(ELISA)after 28 days.Results:The levels of serum NO decreased significantly(P<0.01),and the levels of serum ET-1 increased significantly(P<0.01)in model group compared with sham-operation group;The levels of serum NO increased significantly,and the levels of serum ET-1 decreased significantly(P<0.01)in Chinese herbal medicine group compared with model group.On the 28th day of the drug administration,the levels of serum NO increased significantly,and the levels of serum ET-1 increased(P<0.05)in Chinese herbal medicine group compared with Quinapril group.Conclusion:There is a significant decrease in the levels of serum NO and increase in the levels of serum ET-1 of coronary microcirculation endothelial injury in rats with myocardial infarction.The Chinese medicine group can increase NO and decrease ET-1,and improve the endothelial function of the microvascular lesions;In 28 days,compared wi
