摘  要:目的:通过超滤法对刺五加果多糖的分子量分级,筛选出抗氧化活性较好的分子量组分。方法:采用热水提取醇沉淀法提取刺五加果多糖,三氯乙酸法脱除蛋白质,不同截留分子量的超滤膜对刺五加果多糖进行分离,各级多糖通过测定DPPH和超氧自由基O 2-的清除率来进行筛选其抗氧化活性组分。结果:刺五加果多糖经三氯乙酸除蛋白,超滤法分离后,分子量不同的各组分的多糖含量均相应的增大,50~10kDa组分含量最高为88.15%。通过测定DPPH和超氧自由基O 2-的清除率发现,分子量不同的各组分刺五加果多糖均具有不同程度的抗氧化活性,其中抗氧化活性最好的是分子量50~10kDa组分。结论:通过超滤法可以将刺五加果多糖有效的分离和纯化,并通过体外抗氧化实验筛选出最好的活性组分,达到了分离和筛选的目的。Objective:To isolate and screen the molecular weight components of Acanthopanax polysaccharide with good antioxidant activity.Methods:Acanthopanax fruit polysaccharides were extracted by hot water extraction and alcohol precipitation method.The protein was removed by trichloroacetic acid method,and then ultrafiltration membrane with different molecular weight was used to separate Acanthopanax fruit polysaccharides.The antioxidant active components of polysaccharides at all levels were screened by determination of DPPH and superoxide radical O 2-clearance rate.Results:The polysaccharides of Acanthodichloroacetic acid were removed by ultrafiltration,and the polysaccharides content of each component with different molecular weight increased correspondingly,with the maximum content of 50~10 kDa component being 88.15%.Through the determination of DPPH and superoxide radical O 2-clearance rate,it was found that each component of Acanthopanax polysaccharide with different molecular weight had different antioxidant activity,among which the component with the best antioxidant activity was the component with molecular weight of 50~10 kDa.Conclusion:The polysaccharides of Acanthopanax quinquefolium can be separated and purified effectively by ultrafiltration,and the best active component can be screened out by antioxidant experiment in vitro.
