摘  要:目的:探讨丹皮酚对牙龈卟啉单胞菌的体外抗菌性。方法:在体外常规培育牙龈卟啉单胞菌,并使不同浓度的丹皮酚与其共培养,将1%氯己定作为阳性对照组。通过直接接触法测定丹皮酚在浓度为0、1.5、3、7.5及15 mmol/L时对牙龈卟啉单胞菌的抗菌性;采用菌落形成单位(CFU)检测丹皮酚在浓度为0、1.5、3、7.5、15及20 mmol/L时牙龈卟啉单胞菌的活细菌数;采用活死细菌染色法分析丹皮酚浓度为0、1.5、3、7.5、15及20 mmol/L时牙龈卟啉单胞菌活死细菌的比例。结果:丹皮酚浓度为1.5、3、7.5、15及20 mmol/L时,牙龈卟啉单胞菌的OD600均明显下降,且呈浓度依赖性,差异均有统计学意义(P<0.05)。丹皮酚浓度为3、7.5、15及20 mmol/L时,对牙龈卟啉单胞菌均有一定的抗菌结果,随着丹皮酚浓度的增加,其抗菌效果越佳,且各组间比较差异均有统计学意义(P<0.05)。不同浓度的丹皮酚(3、7.5、15及20 mmol/L)处理牙龈卟啉单胞菌生物膜后,发现空白对照组以绿色荧光为主,红色荧光不明显;而丹皮酚3、7.5、15和20 mmol/L组以及阳性对照组以红色荧光为主,绿色荧光较弱,表明丹皮酚对牙龈卟啉单胞菌生物膜有显著的杀灭作用,且随着浓度从0~15 mmol/L的逐渐增大,其对牙龈卟啉单胞菌生物膜的杀灭作用越强,各组间比较差异均有统计学意义(P<0.05);丹皮酚15 mmol/L与20 mmol/L对牙龈卟啉单胞菌生物膜的杀灭作用相似,组间比较差异无统计学意义(P>0.05)。结论:丹皮酚对牙龈卟啉单胞菌具有较佳的体外抗菌效果,可将其作为治疗慢性牙周炎的药物成分之一,临床应用前景良好。OBJECTIVE:To probe into the in vitro antibacterial effect of paeonol on Porphyromonas gingivalis.METHODS:Porphyromonas gingivalis was routinely cultured in vitro and co-cultured with paeonol at different concentration,1%chlorhexidine was set for positive control.The antibacterial properties of paeonol on Porphyromonas gingivalis were determined by direct contact method at the concentrations of 0,1.5,3,7.5 and 15 mmol/L;the number of viable bacteria of Porphyromonas gingivalis was measured by colony forming unit(CFU)when the concentrations of paeonol were 0,1.5,3,7.5,15 and 20 mmol/L;the proportion of living dead bacteria in Porphyromonas gingivalis was determined by living dead bacteria staining when the concentrations of paeonol were 0,1.5,3,7.5,15 and 20 mmol/L.RESULTS:When the concentrations of paeonol were 1.5,3,7.5,15 and 20 mmol/L,the OD600 of Porphyromonas gingivalis decreased significantly and presented a concentration-dependent,with statistically significant difference(P<0.05).When the concentrations of paeonol were 3,7.5,15 and 20 mmol/L,it had a certain antibacterial effect on Porphyromonas gingivalis,and the antibacterial effect turned better with the increasing of paeonol concentration,and the differences among all groups were statistically significant(P<0.05).After different concentrations of paeonol(3,7.5,15 and 20 mmol/L)were treated with Porphyromonas gingivalis biofilms,it was found that green fluorescence was dominant in the control group,while red fluorescence was not obvious;while in 3,7.5,15 and 20 mmol/L group and positive control group,the red fluorescence was dominant,the green fluorescence was weak,which indicated that paeonol had significant killing effects on Porphyromonas gingivalis biofilms,and as the concentration gradually increases from 0-15 mmol/L,the killing effect turned stronger,the differences among all groups were statistically significant(P<0.05);while the paeonol had similar killing effects on Porphyromonas gingivalis biofilms at the concentration of 15 mmol/L and 20 mmol/
