摘  要:目的利用傅里叶变换红外(Fourier transform infrared,FTIR)显微光谱技术结合化学计量学方法。分析挫伤骨骼肌组织光谱差异性与损伤时间的关系,为损伤时间推断提供新的技术。方法建立大鼠骨骼肌挫伤模型,于损伤后预设的不同时间点将大鼠处死后,并分别于死后0h和死后24h时损伤处取大鼠骨骼肌组织。应用FTIR显微光谱技术进行数据采集获得原始数据后采用随机森林、支持向量机等化学计量学方法进行运算分析。结果大鼠骨骼肌组织光谱吸收峰强度随损伤时间延长发生变化;各损伤时间点光谱有聚类趋势。支持向量机构建的判别模型进行4h^24h和28h^48h二分类内部交叉验证准确率达91.7%,外部验证准确率达83.3%;在二分类基础上随机森林对4h^24h内各时间点的内部验证和外部验证平均准确率分别为83.3%、75.0%。经过对死后0h和死后24h样本分析,同一死亡时间的不同损伤时间光谱亦有明显区分。结论红外光谱结合化学计量学方法可对骨骼肌挫伤组织进行有效分析并建立分类判别模型,对损伤时间推断有参考价值。Objective To analyze the relationship between FTIR spectrum and injury time of skeletal muscle contusion,the Fourier transform infrared(FTIR)microspectroscopy combined with chemometrics was used as a new technique for injury time estimation.Methods The skeletal muscle contusion model of rats were established,then FTIR spectrum data were collected at different interval time points after injury,and the raw data were analyzed by chemometrics.In order to estimate the injury time of samples,the principal component analysis,partial least squares discrimination analysis,support vector machine and random forest classification were performed to analyze the difference of simples.Results The spectral absorption peak intensity of skeletal muscle tissue changed with injury time.The spectral samples at each time point showed clustering trend.The internal verification accuracy and the external verification accuracy of discriminant model constructed by SVM were 91.7%and 83.3%in 4 h-24 h and 28 h-48 h dichotomy,respectively.In addition,the average accuracy of internal and external validation of random forest for each time point within 4 h-24 h was 83.3%and 75.0%,respectively.After analyzing the samples at 0 h and 24 h after death,the different injury time spectra of the same death time were also clearly distinguished.Conclusion FTIR spectroscopy combined with chemometrics can provide effective analysis of contusive skeletal muscle tissue and establish classification model,so as to provide valuable reference for injury time estimation.
