摘 要:信号素3A是一类信号转导蛋白,是一类调节血管生成、分支形态发生、轴突生长和细胞迁移的导向蛋白,在器官发生、免疫应答和肿瘤等方面具有多种调节作用。Sema3A由足细胞分泌,是正常肾脏形态和肾小球滤过屏障发育所必需的。研究发现,正常发育的肾脏中足细胞Sema3A的过表达会导致小鼠蛋白尿性肾小球疾病。体内Sema3A过表达可导致足突消失、肾小球基底层分层和内皮细胞损伤,并通过下调去足细胞去氧肾上腺素(nephrin)和抑制αvβ3整合素从而破坏足细胞形态细胞。去足细胞去氧肾上腺素与Sema3A的受体神经丛蛋白A1之间有一种新的直接相互作用。去足细胞去氧肾上腺素-神经丛蛋白A1相互作用使足细胞裂孔隔膜(slit-diaphragm)信号复合体与细胞外Sema3A信号连接起来起作用。因此,Sema3A作为细胞外负调节因子对肾小球滤过屏障结构和功能发挥着重要作用。Semaphore 3a is a type of signal transduction protein,a type of guiding protein that regulates angiogenesis,branch morphogenesis,axon growth and cell migration,and has a variety of regulatory effects in organogenesis,immune response and tumors.Sema 3A is secreted by podocytes and is required for normal kidney morphology and glomerular filtration barrier development.Studies have found that overexpression of Sema 3A in podocytes in normally developing kidneys can cause proteinuria glomerulopathy in mice.Sema 3A overexpression in vivo can lead to the disappearance of foot processes,glomerular basal layer delamination and endothelial cell damage,and it can destroy podocyte morphology cells by down-regulating depoedocyte nephrin(nephrin)and inhibitingαvβ3 integrin.There is a new direct interaction between Phenylephrine and Sema3A receptor plexin A1.The depoedocyte phenylephrine-plexin A1 interaction connects the podocyte slit-diaphragm signaling complex with extracellular Sema3A signaling.Therefore,Sema3A,as an extracellular negative regulator,plays an important role in the structure and function of the glomerular filtration barrier.
