摘 要:为探讨矮小型品系S3系和苏禽3号不同等级卵泡的膜层和颗粒层中生长分化因子9(Growth differentiation factor9,GDF9)的表达差异和表达模式,分别于24和43周龄采集两个品系母鸡的大白卵泡(Large white follicle,LWF,4~6 mm)、小黄卵泡(Small yellow follicle,SYF,6~8 mm)以及等级卵泡F1、F2、F3、F4、F5的膜层和颗粒层,每个组织5个重复;采取荧光定量PCR法检测两个品种各级卵泡中GDF9 mRNA的表达量。结果显示:GDF9 mRNA在2个鸡种卵泡膜层和颗粒层的表达均呈"低-高-低"趋势,表达高峰在SYF期,且明显高于其他卵泡期(P<0.05),在F1和F2期几乎不表达;同一等级卵泡的膜层和颗粒层在不同时期表达模式十分相似,均为24周龄高于43周龄(P>0.05);不同品种鸡卵泡颗粒层和膜层GDF9 mRNA的表达规律为苏禽3号略高于S3系(P>0.05)。研究结果提示,GDF9基因在鸡卵泡中的表达存在组织、时间及品种差异,GDF9主要影响鸡的分级前卵泡,其高表达可能对维持大量卵泡生长有重要作用。This study investigated the expression differences and expression patterns of growth differentiation factor 9(GDF9) in the membrane layer and particle layer of different grades of follicles of dwarf chicken S3 line and Suqin No.3. Large white follicle(LWF, 4 to 6 mm), small yellow follicle(SYF, 6 to 8 mm) and grade follicle(F1, F2, F3, F4,and F5) were collected at 24 and 43 weeks of age, respectively. The mRNA expression of GDF9 gene in the follicular tissues of the two breeds was detected by fluorescence quantitative PCR. The results showed that the expression of GDF9 mRNA in the membrane layer and particle layer of follicles showed a trend of "low-high-low" with the continuous development of follicles, and the expression reached the peak in SYF, which was signifcantly higher than in the other follicles(P<0.05);GDF9 mRNA was nearly no expression in F2 and F1;The expression of GDF9 mRNA in the granular layer and membranous layer of the same grade follicles at 24 weeks of age was higher than that of 43 weeks of age(P>0.05), which in granular layer and membranous layer of all follicles of Suqin No.3 was higher than those of S3 line(P>0.05). It’s indicated that there were differences in tissue, time and line in the expression of GDF9 gene in chicken follicles, and GDF9 mainly affected the pre-hierarchal follicle in chicken, and its high expression may play an important role in growth maintaining for large number of follicles.
