摘  要:[目的]本文旨在研究脱氢表雄酮(DHEA)对高脂日粮饲喂大鼠脂肪组织炎症以及脂质沉积的影响,为后期探讨DHEA调控机体脂代谢紊乱及其炎症反应相关机制的研究提供理论基础。[方法]采用正常日粮(ND)、高脂日粮(HFD)和高脂日粮+DHEA(HFD+DHEA)连续饲喂雄性Sprague-Dawley(SD)大鼠(3组,每组10只)10周,采集大鼠不同组织部位的脂肪组织进行苏木素伊红(HE)染色,分析脂肪细胞大小,荧光定量PCR法检测脂肪代谢及炎症因子相关基因表达水平。[结果]与正常日粮组相比,高脂日粮组大鼠脂肪细胞大小极显著增加(P<0.01),脂肪组织中乙酰辅酶A羧化酶(ACC)、脂肪合成酶(FAS)、固醇调节元件结合蛋白1c(SREBP-1c)、白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α)基因mRNA表达水平均显著上调(P<0.05);同时肉毒碱棕榈酰转移酶1(CPT-1)、肉毒碱棕榈酰转移酶2(CPT-2)和过氧化物酶体增殖物激活受体γ共激活因子1α(PGC-1α)基因mRNA表达水平均显著下调(P<0.05)。与高脂日粮组相比,DHEA处理可显著抑制高脂日粮饲喂大鼠的不同组织部位脂肪细胞大小的增加(P<0.05)。与高脂日粮组相比,DHEA处理显著减少高脂日粮饲喂大鼠的脂肪合成相关因子和促炎细胞因子的表达水平,同时显著增加脂肪分解相关因子的表达水平(P<0.05)。[结论]DHEA能缓解高脂日粮诱导的脂肪组织中的炎症反应,并减少脂肪细胞中脂质的沉积。[Objectives]The present study aimed to investigate the effects of dehydroepiandrosterone(DHEA)on adipose tissue inflammation and lipid accumulation in rats fed with high fat diet,to provide a basis for studying the mechanism of DHEA regulating lipid metabolism and inflammation response.[Methods]Male Sprague-Dawley(SD)rats(three groups,10 in each group)were fed with normal diet(ND),high fat diet(HFD)and high fat diet with dehydroepiandrosterone(HFD+DHEA)for 10 weeks.After that,rat adipose tissue was collected for hematoxylin-eosin(HE)staining to analyze the sizes of adipocytes.The expression levels of genes related to inflammation and lipid metabolism were detected by qPCR.[Results]Compared with the normal diet group,the adipocytes sizes significantly increased in rats fed a high fat diet(P<0.01).The mRNA expression levels of interleukin-6(IL-6),tumor necrosis factor alpha(TNF-α),acetyl-CoA carboxylase(ACC),fatty acid synthase(FAS)and sterol regulatory element binding protein-1c(SREBP-1c)significantly increased(P<0.05),while the mRAN expression levels of carnitine palmitoyltransferase-1(CPT-1),carnitine palmitoyltransferase-2(CPT-2)and peroxisome proliferator-activated receptor-γcoactivator-1α(PGC-1α)significantly decreased in rats fed a high fat diet(P<0.05).Compared with the high fat diet group,the increasing of adipocytes sizes induced by high fat diet were significantly inhibited after DHEA treatment(P<0.05);DHEA treatment also significantly decreased the expression levels of pro-inflammatory and lipogenesis-related factors,while the expression levels of lipolysis-related factors significantly increased(P<0.05).[Conclusions]Dehydroepiandrosterone treatment can alleviate the inflammatory response in adipose tissue caused by a high fat diet and reduce the lipids accumulation in adipocytes.
