基于生物信息学分析法探讨BIRC5(Survivin)在肾透明细胞癌中的表达及其与预后的关系

摘　　要：目的探讨BIRC5在肾透明细胞癌肿瘤组织中的表达水平及其与临床预后之间的关系。方法首先使用Oncomine以及癌症基因组图谱数据库预测BIRC5 mRNA在肾透明细胞癌中的表达情况,并探索其表达量与生存的关系。采用生存分析和比例风险回归模型评估BIRC5表达与肾透明细胞癌患者预后的相关性,并通过基因集变异分析以及基因集富集分析探讨BIRC5所富集的相关通路。此外,探究了BIRC5在肾透明细胞癌中共表达基因的情况。结果肾透明细胞癌组织中BIRC5 mRNA的表达水平明显高于正常组织(P<0.001)。生存分析显示,BIRC5高表达患者的生存时间显著减少(P<0.001)。此外,还发现BIRC5差异表达水平与病理分级、临床TNM分期以及性别相关(P均<0.01)。单因素、多因素比例风险回归模型表明BIRC5表达是肾透明细胞癌患者生存时间的独立预后因素(P均<0.05)。结论BIRC5在肾透明细胞癌中高表达,是影响预后的重要因素。BIRC5可能是肾透明细胞癌潜在的预后生物学标志物及治疗靶标。Objective To investigate the expression of BIRC5 in clear cell renal cell carcinoma(ccRCC)and its association with prognosis.Methods The mRNA expression of BIRC5 in ccRCC was analyzed in Oncomine and TCGA databases,and its association with ccRCC prognosis was predicted.The correlation between BIRC5 expression and ccRCC prognosis was evaluated with Kaplan-Meier analysis and proportional hazard regression model.The related genes and pathways were obtained from TCGA database with gene set enrichment analysis(GSEA)and gene set variation analysis(GSVA).The co-expression of BIRC5 gene was investigated.Results The mRNA expression of BIRC5 was significantly higher in ccRCC tissues than in normal tissues(P<0.001).Kaplan-Meier analysis showed the overall survival(OS)of ccRCC patients with high BIRC5 expression was significantly reduced(P<0.001).The differential expression of BIRC5 was correlated with pathological grade,clinical TNM stage and gender(all P<0.05).Univariate and multivariate Cox analyses showed the expression of BIRC5 was an independent prognostic factor for the survival of ccRCC patients.Conclusion BIRC5 is highly expressed in ccRCC and is an important factor affecting prognosis.BIRC5 may be a potential prognostic biomarker and therapeutic target for ccRCC.
