摘　　要：目的通过灌胃不同煎煮时间的熟大黄水煎液治疗慢性肾功能衰竭(chronic renal failure,CRF)大鼠,观察CRF大鼠肾功能改善情况,探讨熟大黄治疗CRF大鼠的最佳煎煮时间。方法将CRF大鼠模型分别用熟大黄水煎液(煎煮5、7、9、10、15、20、30 min)灌胃治疗,测定血BUN、Cr、UA值,免疫组化、免疫印迹试验检测Akt、PI3k、TGF-β1等蛋白在熟大黄治疗后的CRF大鼠肾组织表达水平。结果与模型组相比,熟大黄治疗组血BUN、Cr、UA值均显著下降(P<0.01),大鼠肾组织中Akt、PI3k、TGF-β1的平均光密度比和灰度比值均显著降低(P<0.01),其中熟大黄10 min组下降最明显(P<0.05)。结论熟大黄后下煎煮10 min更适用于CRF大鼠的治疗。Objective To investigate the most effective decocting time of Shudahuang(Prepared Rhubarb)in chronic renal failure(CRF)rats by giving CRF rats decoction of Shudahuang(Prepared Rhubarb)at different decocting time by gavage.Methods Rat models of CRF were treated with decoction of Shudahuang(Prepared Rhubarb)(decocting for 5,7,9,10,15,20,30 minutes)by gavage.The serum levels of BUN,Cr and UA were measured.IHC and WB were used to detect the effects on the expressions of TGF-β1,PI3k and Akt in CRF rats.Results Compared with those of the model group,the serum levels of BUN,Cr and UA,the mean optical density ratio and gray scale ratio of TGF-β1,PI3k and Akt in kidney tissue of rats decreased significantly(P<0.01)and Shudahuang(Prepared Rhubarb)with a decocting time of about 10 minutes group decreased significantly(P<0.05).Conclusion Shudahuang(Prepared Rhubarb)with decocting time of 10 minutes after boiling is more appropriate for CRF rats treatment.
