摘 要:目的探讨肾舒胶囊对2型糖尿病肾病大鼠的治疗作用以及对AKT/NF-κB信号通路的影响。方法将雄性SD大鼠随机分为4组(n=10),即正常组、模型组、肾舒胶囊低剂量组、肾舒胶囊高剂量组,对照组予以正常饲料喂养,造模组予以高糖高脂饲料(在普通饲料的基础上加入10%的白砂糖和20%的猪油)喂养2个月后,予以腹腔注射链脲佐菌素(streptozoticin,STZ)35 mg/kg并诱导糖尿病肾病模型,造模成功后,治疗组分别予以肾舒胶囊[0.018 g/(kg·d),0.3 g/(kg·d)]灌胃。连续给药1周后检测大鼠体质量、血清肌酐、血清尿素氮、24 h尿蛋白等指标;HE染色观察肾脏病理变化,PAS染色观察肾脏组织中的糖原和黏液变化;采用Western Blot检测各组大鼠肾脏组织AKT、p-AKT、NF-κB(p65)、p-NF-κB(p-p65)、α-SMA的表达水平。结果与模型组相比,肾舒胶囊可显著改善糖尿病肾病大鼠肾功能、24 h尿蛋白定量和肾脏病理,下调肾脏组织中p-AKT、p-NF-κB p 65、α-SMA的表达。结论肾舒胶囊可改善糖尿病大鼠肾脏损害,其机制可能与抑制AKT/NF-κB信号通路改善纤维化水平有关。Objective To investigate the therapeutic effect of Shenshu Capsule on type 2 diabetic nephropathy rats and its effect on AKT/NF-κB signaling pathway.Methods Male SD rats were randomly divided into 4 groups(n=10),namely the normal group,the model group,the Shenshu Capsule low-dose groupand the Shenshu Capsule high-dose group.The control group was fed with normal feed.High-sugar and high-fat feed(adding 10%white granulated sugar and 20%lard on the basis of ordinary feed)was given for modeling.After feeding for 2 months,intraperitoneal injection of streptozoticin(STZ)35 mg/kginduced the model of diabetic nephropathy.The treatment group was orally administered Shenshu Capsules[0.018 g/(kg·d),0.3 g/(kg·d)].After 1 week of continuous administration,body weight,serum creatinine,serum urea nitrogenand 24-hour urine protein were measured.HE staining was used to observe renal pathological changes,and PAS staining was used to observe changes in glycogen and mucus in kidney tissues.The expression levels of AKT,p-AKT,NF-κB(p65),p-NF-κB(p-p65)andα-SMA in kidney tissue were compared.Results Compared with the model group,Shenshu Capsule can significantly improve renal function,24-hour urine protein quantification and renal pathology in diabetic nephropathy rats,and down-regulate expressions of p-AKT,p-NF-κB p65 andα-SMA.Conclusion Shenshu Capsule can improve kidney damage in diabetic rats,and its mechanism may be related to inhibition of AKT/NF-κB signaling pathway and improvement of fibrosis.
