过表达GA调控相关基因SmGASA4提高拟南芥抗逆性的研究

摘　　要：为了研究赤霉素(gibberellins,GAs)调控相关基因SmGASA4在植物的生长发育和应对外界胁迫方面的作用,本实验根据前期丹参根转录组数据库获得一个与GA调控相关的基因SmGASA4,构建过表达载体转染拟南芥,并对过表达SmGASA4拟南芥表型进行观察,分别用干旱、200mmol/L NaCl、200μmol/L GA3、50μmol/L多效唑胁迫处理.在正常生长条件下,过表达植株叶片长度是野生型的1.21倍果荚长度是1.25倍;干旱处理过表达拟南芥株高是野生型的1.54倍,果荚数量是2.46倍;过表达植株的抗盐能力要优于野生型;GA3处理过表达拟南芥株高是野生型的2.63倍果荚数量是9.17倍,对于阐明SmGASA4的抗逆功能具有重要意义.To study the gibberellin(gibberellins, GAs) regulate gene Sm GASA4 function role in plant growth and response to external stress, This experiment get one GA regulate gene of SmGASA4 from Salvia miltiorrhiza according to previous transcriptome data and build vector. Getting SmGASA4 over expression material of Arabidopsis, and observation the phenotypic of over expression Sm GASA4 in Arabidopsis. Taking drought, 200 mmol/L salt, 200 μmol/L GA3, 50 μmol/L paclobutrazol stress treatment.Under normal growth condition, overexpression 1.21 times of plant leaf length than wild type,Silique length is 1.25 times;Drought stress over expression Arabidopsis plant height 1.54 times than wild type, Silique number is 2.46 times;Overexpression of Sm GASA4 Arabidopsis have a strong resistance of salt than wild type;GA3 process over expression Arabidopsis plant height 2.63 times than wild type, Silique number is 9.17 times;this study have great meaning in explain the SmGASA4 function in stress resistance.
