摘  要:为探讨蒙古口蘑(Tricholoma mongolicum)子实体不同极性提取物对免疫抑制小鼠免疫功能的影响,依次用石油醚、乙酸乙酯、水提取子实体得到3种提取物;将小鼠随机分为空白组、模型组及石油醚、乙酸乙酯和水的提取物高中低剂量组,检测小鼠巨噬细胞的吞噬功能,测定溶血素生成量和血清中白细胞介素-2(interleukin-2,IL-2)、IL-4、干扰素-γ(interferon-γ,IFN-γ)、免疫球蛋白A(immunoglobulin A,IgA)含量,计算胸腺和脾脏指数,分别观察胸腺和脾脏组织的苏木精-伊红和免疫组织化学染色切片。结果表明:与模型组相比,石油醚提取物、乙酸乙酯提取物和水提物均可以增加小鼠体质量,提高胸腺和脾脏指数;石油醚提取物高剂量组和乙酸乙酯提取物中剂量组可提高巨噬细胞吞噬百分率和吞噬指数及溶血素生成量,提高细胞因子IL-2、IL-4、IFN-γ和IgA的水平,明显改善免疫抑制小鼠胸腺和脾脏组织的病变情况,石油醚提取物各剂量组的各指标恢复程度优于其他提取物组。Fruiting bodies of Tricholoma mongolicum were extracted in order by petroleum ether,ethyl acetate and water to yield extracts with different polarities.These extracts were then used to treat immunocompromised mice to study their effects on immune function in mice.Mice were randomly divided equally into 12 groups as follows:control group,model group,positive control group,petroleum ether extract groups(high,medium and low doses),ethyl acetate extract groups(high,medium and low doses),and water extract groups(high,medium and low doses).The immunocompromised model was established by administration of 60 mg·kg^-1 cyclophosphamide(CTX)intraperitoneally at day 1,2,3 and 6,respectively.Levamisole hydrochloride(LM)was administered intragastrically to immunocompromised mice at 50 mg·kg^-1 everyday to serve as the positive control.For mice treated by different doses of petroleum ether extract,they were administered intragastrically with high(30 mg·kg^-1),medium(20 mg·kg^-1)and low(10 mg·kg^-1)doses for 14 d consecutively,respectively.For mice treated by different doses of ethyl acetate extract,they were administered intragastrically with high(30 mg·kg^-1),medium(20 mg·kg^-1)and low(10 mg·kg^-1)doses for 14 d consecutively,respectively.For mice treated by different doses of water extract,they were administered intragastrically with high(1000 mg·kg^-1),medium(500 mg·kg^-1)and low(250 mg·kg^-1)doses for 14 d consecutively,respectively.Macrophage phagocytosis,thymus index,spleen index and serum levels of IL-2,IL-4,IFN-γ,IgA and hemolysin were measured for mice in each group.Tissue morphology of thymus and spleen was examined by hematoxylin-eosin(HE)staining.Levels of F4/80 protein in thymus and spleen were also observed by immunohistochemistry.The results showed that all three kinds of extracts improved body weight,thymus index and spleen index compared with the model group.In addition,high dose petrolum ether extract and medium dose ethyl acetate extract increased phagocytosis percentage,phagocytic index,and serum
