摘 要:目的进行1例疑似AB3亚型的献血者血型分子生物学研究。方法用常规血型血清学方法进行ABO血型鉴定;对ABO基因的外显子6和外显子7扩增后进行直接测序和单克隆测序;通过短串联重复序列检测分析14个基因位点。结果该献血者血清学表现为AB3亚型,而ABO基因分型、序列测定以及STR-PCR检测的多个位点均存在2个以上的等位基因。结论该献血者非AB3亚型,而是罕见的卡米拉血型。Objective To perform the molecular analysis of a case initially suspected as AB3 subtype and actually a rare Chimeras blood group.Methods ABO blood group of this blood donor was identified by routine serological test. Exon 6 and exon 7 of ABO genes were subjected to direct sequencing and monocloned sequencing to determine the genotypes, and 14 loci were studied by short tandem repeat(STR) sequence analysis.Results The blood type serological testing of this blood donor presented as AB3 phenotype, while more than 2 alleles at multiple loci were detected by ABO genotyping, sequencing and STR analysis.Conclusion The rare Chimeras blood type, though suspected as AB3 initially, was revealed by DNA sequencing.
