摘  要:为探究GLIS1基因表达水平与绵羊产羔数之间的关系,利用半定量RT-PCR(Semi-quantitative RT-PCR,sqRT-PCR)和实时荧光定量PCR(Quantitative real-time PCR,qPCR)技术检测了绵羊GLIS1基因在不同产羔能力绵羊各组织中的表达情况。结果表明:GLIS1基因在多羔小尾寒羊输卵管高表达,在单羔苏尼特羊卵巢、输卵管、子宫体以及子宫角中的表达量均高于多羔小尾寒羊(P>0.05);GLIS1基因B+基因型在各组织中的表达量均极显著高于BB基因型和++基因型(P<0.01)。综上得出,GLIS1基因可能通过调控其mRNA表达水平对绵羊产羔数发挥负调控作用。To investigate the association between expression level and litter size in sheep,semi-quantitative RT-PCR(sqRT-PCR)and quantitative real-time PCR(qPCR)were performed to detect the expression of GLIS1 gene in sheep with different lambing capacity.The results showed that the expression of GLIS1 was at high level in the oviduct of Small Tail Han sheep,the expression in ovary,oviduct,uterus body and uterine horn of monotocous Sunite sheep was higher than that in polytocous Small Tail Han sheep;the expression of GLIS1 in B+was extremely significant higher than that in BB and++(P<0.01).Therefore,it can be concluded that GLIS1 gene may play a negative regulatory role in the litter size of sheep by regulating its mRNA expression level.
