摘  要:为了解样本溶血对血凝抑制(HI)试验和胶体金法检测新城疫病毒抗体的影响,通过冻融方法、常规方法和模拟方法制备了不同程度的溶血样本,应用血凝抑制试验和胶体金法检测了样本中新城疫病毒抗体.结果表明:模拟溶血样本中,5%~45%溶血对HI试验结果无影响,但50%溶血样本的HI效价降低了;冻融方法制备的溶血样本溶血程度远高于模拟的50%溶血样本,总体水平上冻融溶血样本的HI效价显著降低了;不同程度溶血的检测样本几乎不影响HI试验和胶体金法的检测结果.这说明在应用HI方法和胶体金法进行新城疫病毒抗体检测时,样本溶血对结果判定没有显著影响,但高度溶血样本可能会影响HI抗体效价,不能使用冻融方法制备待检血清.In order to understand the effect of sample hemolysis on the detection of Newcastle disease virus antibody by hemagglutination inhibition(HI)test and colloidal gold method,the present study prepared different degrees of hemolysis samples by freeze-thaw method,conventional method and simulation method.The antibody of Newcastle disease virus in prepared samples was detected by HI test and colloidal gold method.The results showed that in the simulated hemolysis samples,5%-45%hemolysis had no effect on the results of HI test,but the HI titer of 50%hemolysis samples was reduced.The hemolysis degree of samples prepared by freeze-thaw method was much higher than that of the simulated 50%hemolysis samples,and the HI titer of the freeze-thaw hemolysis samples was significantly reduced at the overall level.The determination of HI test and colloidal gold test was almost not affected by the hemolysis of the samples.These indicates that when using HI method and colloidal gold method for the detection of Newcastle disease virus antibody,the hemolysis of samples have no significant effect on the determination of the results,but the high degree of hemolysis samples may affect the HI antibody titer,and the freeze-thaw method cannot be used to prepare the serum for test.
