摘　　要：为研究奶牛亚临床型和临床型乳房炎乳腺病理组织学的变化,采用石蜡切片法和苏木精-伊红染色法(HE染色法)进行病理组织学观察;采用甲苯胺蓝染色法(RTB)和改良甲苯胺蓝染色法(MRTB)染色对比观察组织中肥大细胞染色效果、形态和数量变化。结果表明,随着乳腺组织炎症的加剧,乳头管表面角层脱落加剧,腺管、腺泡上皮坏死、脱落,炎性细胞及肥大细胞发生浸润,潜在性的炎症转变为临床型乳房炎。非临床型乳房炎乳腺组织的肥大细胞多分布在腺小叶、腺泡间靠近基膜处及小叶间结缔组织的血管附近,细胞呈圆形、椭圆形或梭形。临床型乳房炎的肥大细胞较多分布在乳头管、腺管之间及小叶间的血管附近,数量较多,细胞呈圆形或椭圆形。对比染色效果、阳性细胞数目,改良甲苯胺蓝染色法对乳腺组织中肥大细胞的显示更清晰、准确,且能更长时间保存。To study histopathological changes of mammary gland with subclinical and clinical mastitis in dairy cows,the paraffin section method and hematoxylin-eosin staining method were used for histopathological observation.Routine toluidine blue staining and the modified routine toluidine blue staining(MRTB)were used to observe the staining effects,morphology and number of mast cells in tissues.The results showed that with inflammation of mammary glands intensifies,the keratin on the surface of the nipple tube decreased.The acinar duct and epithelium show necrosis and shedding;the inflammatory cells and mast cells infiltrate in the tissue;the latent inflammation turnes into clinical mastitis.The mast cells of the mammary gland tissue of subclinical mastitis are mostly distributed in the blood vessels of the glandular lobules near the basal membrane and the interlobular connective tissue,and the cells are round,oval or fusiform.The mast cells of clinical mastitis are mostly distributed in the vicinity of the blood vessels between the papillary duct,glandular duct and lobules,and the cells are round or oval.By comparing the staining effect and the number of positive cells,the MRTB showed a clearer and more accurate indicator of mast cells in the mammary gland tissue and could be stored for a longer time.
