摘　　要：目的观察维生素D3联合化疗对结核病小鼠T淋巴细胞亚群内穿孔素和颗粒溶素表达的影响,为临床应用维生素D3进行结核病的宿主导向治疗提供实验数据支持。方法通过尾静脉注射结核分枝杆菌构建小鼠结核病模型,并使用化疗药、维生素D3以及化疗药联合维生素D3进行干预治疗。流式细胞术检测小鼠外周血中CD4+和CD8+T淋巴细胞中穿孔素和颗粒溶素的表达水平,ELISA检测小鼠血清中IL-10和IL-17A水平。结果⑴维生素D3辅助化疗能够显著降低小鼠外周血T淋巴细胞中总穿孔素和颗粒溶素以及CD4+和CD8+T淋巴细胞中穿孔素和颗粒溶素的表达(P<0.05);⑵维生素D3辅助化疗能够显著升高结核病小鼠血清IL-10含量,并显著抑制IL-17A水平。结论维生素D3辅助化疗可以通过在疾病活跃期调节细胞毒性分子的表达来帮助宿主控制炎症和组织损伤,从而有助于缩短宿主治疗时间。Objective To observe the effect of vitamin D3 combined with chemotherapy on the expression of perforin and granlysin in T-lymphocyte subsets of mice with tuberculosis,and to provide experimental data support for clinical application of vitamin D3 in host directed treatment of tuberculosis.Methods The mouse model of tuberculosis was constructed by injecting Mycobacterium tuberculosis through the tail vein,and intervention therapy was performed using chemotherapy drugs,vitamin D3,and chemotherapy drugs combined with vitamin D3.Flow cytometry was used to detect the expression levels of perforin and granulysin in CD4+and CD8+T lymphocytes in the peripheral blood of mice.ELISA was used to detect the levels of IL-10 and IL-17A in the serum of mice.Results⑴Vitamin D3 adjuvant chemotherapy can significantly reduce the expression of perforin and granulysin in peripheral blood T lymphocytes and CD4+and CD8+T lymphocytes(P<0.05).⑵Vitamin D3 adjuvant chemotherapy can significantly increase the serum IL-10 content of tuberculosis mice,and significantly inhibit IL-17A levels.Conclusion Vitamin D3 adjuvant chemotherapy can help the host to control inflammation and tissue damage by regulating the expression of cytotoxic molecules during the active stage of the disease,thereby helping to shorten the host's treatment time.
