摘  要:细胞发酵罐传统细胞观察方式是人工取出来再放置在倒置目视显微镜下观察,由于现有的倒置目视显微镜工作距较小,无法对罐中状况细节进行实时观察,并且在取液过程中极易造成发酵罐内溶液污染,浪费资源。结合视频显微镜的特点,设计一款用于3~8 L左右的不同曲率细胞发酵罐外部观察的长工作距视频显微镜,显微镜工作距离38 mm,光学放大倍率为-5倍,物方线视场1.2 mm,分辨率δ=2.2μm,使用1/3英寸CCD,并配以光源在发酵罐另一侧进行照射,实现在细胞发酵罐外部进行细胞观察。光学传递函数(MTF)曲线与理论衍射极限接近,达到指标要求,并且分析不同曲率的发酵罐是否对结果有影响进行对比。最后,制造出视频显微镜的原型机,并通过实验对其显示效果进行验证。The traditional cell observation method of the cell fermenter was manually taken out and placed under an inverted visual microscope.Due to the small working distance of the existing inverted visual microscope,the details of the condition in the tank couldn’t be observed in real time,and during the liquid taking process.It was easy to cause solution contamination in the fermenter and waste resources.In this paper,combined with the characteristics of video microscope,a long working distance video microscope for external observation of different curvature cell fermenters of about 3L-8L was designed.The working distance of the microscope was 38mm;the optical magnification was-5X;and the object square field of view was 1.2mm;resolution wasδ=2.2μm;a 1/3-inch CCD was used and with a light source on the other side of the fermenter to achieve cell observation outside the cell fermenter.The optical transfer function(MTF)curve was close to the theoretical diffraction limit,meets the index requirements;and it was analyzed whether fermenters of different curvatures have an effect on the results.Finally,a prototype of the video microscope was fabricated and verified by experiments.
