摘 要:目的探究贯叶连翘提取物对硝酸甘油所致大鼠急性偏头痛的改善作用及其机制。方法将48只SD雄性大鼠用随机数字表法分为正常组、模型组、佐米曲普坦组及贯叶连翘高、中、低3个剂量组,每组8只,后5组大鼠通过从后颈部皮下注射硝酸甘油10 mg/kg建立急性偏头痛大鼠模型。造模成功30 min后分别灌胃给予相应的药物,灌胃后2 h注射10%水合氯醛0.4 ml/100 g麻醉,获取血浆及脑组织,采用ELISA法检测血浆5-羟色胺(5-HT)、多巴胺(DA)、内皮素(ET)和脑组织前列腺素E2(PGE2)水平并作比较和分析。结果与正常组大鼠相比,模型组大鼠血浆5-HT、DA、ET水平和脑组织PGE2水平均显著降低(均P<0.05),说明急性偏头痛大鼠模型建立成功;与模型组相比,各给药组(佐米曲普坦组与贯叶连翘高、中、低组)血浆中5-HT、DA、ET水平和脑组织中PGE2水平均显著增高(均P<0.05),贯叶连翘提取物各剂量组与佐米曲普坦组的作用效果一致。结论贯叶连翘提取物对大鼠体内5-HT、DA、ET、PGE2水平具有一定的调节作用,并对硝酸甘油所致大鼠急性偏头痛有一定的改善作用。Objective To investigate the effects of hypericum perforatum extract on biochemical indicators associated with migraine headache in acute migraine rat model.Methods Forty eight SD male rats were randomly divided into 6 groups with 8 rats in each group:the normal group,model group,zolmittriptan,Hypericum perforatum high,medium and low dose groups.The rat model of acute migraine was established by subcutaneous injection of nitroglycerin(10 mg/kg)in the posterior neck.After 30 min of successful modeling,the corresponding drugs were given by intragastric administration respectively.After 2 h of intragastric administration,10%chloral hydrate(0.4 ml/100 g)was injected to obtain plasma and brain tissue,ELISA was used to detect 5-HT,DA,ET in plasma and the PGE2 in brain tissue.Results Compared to the normal group,the contents of 5-HT,DA,ET in plasma and the PGE2 in brain tissue of the model group were significantly decreased(all P<0.05),indicating the establishment of the rat models of acute migraine was successful.Compared to the model group,the contents of 5-HT,DA and ET in plasma and the content of PGE2 in brain tissue in each treatment group(zolmittriptan,Hypericum perforatum high,medium and low dose groups)were significantly increased(all P<0.05),and the effects of each treatment group were consistent with that of the zolmittriptan group.Conclusion Hypericum perforatum extract can regulate the contents of 5-HT,DA,ET and PGE2 in rats and ameliorate the acute migraine induced by nitroglycerin.
