摘 要:目的观察秦巴硒菇提取物酸性RNA蛋白复合物FA-2-b-β对伯基特淋巴瘤细胞株Raji增殖及凋亡的影响,并探讨其相关作用机制。方法体外培养Raji细胞,分别加入浓度为0.9、1.5、2.1、2.7 mg/mL的FA-2-b-β,对照组加入等量培养基。培养24、48、72 h后收集各组细胞,采用CCK-8法检测FA-2-b-β对Raji细胞的增殖抑制作用;流式细胞仪检测细胞凋亡率;Western blot法检测细胞β-catenin、c-myc、cyclin D1、Bcl-2、Bax蛋白的表达。结果FA-2-b-β能抑制Raji细胞增殖,且呈浓度-时间依赖性(P<0.01);给药48 h后,各浓度FA-2-b-β组细胞凋亡率较对照组均显著上升,且呈剂量依赖性(P<0.05);各浓度FA-2-b-β组细胞β-catenin、c-myc、cyclin D1、Bcl-2蛋白的表达水平显著低于对照组,且呈剂量依赖性(P<0.05),Bax蛋白表达水平显著高于对照组,同样呈剂量依赖性(P<0.05),Bax/Bcl-2比值增加。结论FA-2-b-β呈时间-剂量依赖性诱导伯基特淋巴瘤凋亡,可能与Wnt/β-catenin信号通路相关。Objective To observe the effect of acidic RNA protein complex FA-2-b-β,an extract from Agaricus blazei Murrill-Qbsg,on the proliferation and apoptosis of Burjit lymphoma and investigate the relevant action mechanism.Methods Raji cells were cultured in vitro and added with FA-2-b-βat concentrations of 0.9,1.5,2.1 and 2.7 mg/mL respectively,using those added with the medium at an equal volume as control.The proliferation of Raji cells after culture for 24,48 and 72 h were determined by CCK-8 method,while the apoptosis rate by flow cytometry.The expression levels ofβ-catenin,c-myc,cyclin D1,Bcl-2 and Bax proteins were determined by Western blot.Results FA-2-b-βinhibited the proliferation of Raji cells in a dose-and time-dependent manner(P<0.01).The apoptosis rates of cells treated with FA-2-b-βfor 48 h increased significantly in a dose-dependent manner as compared with that in control group(P<0.05).The expression levels ofβ-catenin,c-myc,cyclin D1 and Bcl-2 in the cells treated with FA-2-b-βat various concentrations were significantly lower,while that of Bax protein was significantly higher,than that in control group(each P<0.05),both in dose-dependent manners.However,the Bax/Bcl-2 ratio increased as compared with that in control group.Conclusion FA-2-b-βinduces the apoptosis of Burkitt lymphoma in a time-and dose-dependent manner by a mechanism which might be associated with Wnt/β-catenin signaling pathway.
