摘 要:目的探究艾滋病病毒(HIV)感染者免疫细胞中NKG2D全长型(NKG2DFL)和截短型(NKG2DTR)两种剪接型含量及其与HIV感染疾病进展的关系,寻找影响NKG2D剪接型含量的可能因素。方法利用实时荧光定量聚合酶链反应(PCR)技术,分别检测HIV慢性感染者与抗病毒治疗后感染者及正常人的不同免疫细胞中NKG2D剪接型含量。在体外用TGF-β刺激后再检测外周血单个核细胞(PBMC)中的NKG2D剪接型含量。结果HIV慢性感染者PBMC中NKG2DFL、NKG2DTR的含量均低于抗病毒治疗后患者(P<0.01)及健康对照(P<0.01),且其NKG2DFL含量与CD4+T淋巴细胞(简称CD4细胞)数目呈明显负相关(r=-0.8405,P<0.01);CD8+T淋巴细胞(简称CD8细胞)和NK细胞中NKG2DFL、NKG2DTR的含量变化与PBMC一致,即HIV慢性感染者中含量低于正常对照(P<0.05),但CD4细胞中NKG2DFL、NKG2DTR含量在HIV慢性感染者反而高于正常对照(P<0.05);经TGF-β刺激后,PBMC中NKG2DFL、NKG2DTR含量均降低。结论HIV慢性感染者中NKG2DFL、NKG2DTR含量均较正常对照降低,尤其是在CD8、NK细胞中;TGF-β使PBMC中NKG2D两种剪接型含量下降。Objective To investigate the expression level of full-length(NKG2DFL)and truncated(NKG2DTR)NKG2D splicing isoforms in immune cells of HIV-infected patients and their relationship with HIV disease progression,so as to find possible factors affecting the splicing isoforms of NKG2D.Methods Real-time quantitative PCR was used to detect the NK-G2D splicing isoforms in different immune cells of the chronically HIV-infected patients,patients treated with antiretroviral therapy and normal controls.The NKG2D splicing isoforms in PBMC were detected after stimulation with TGF-βin vitro.Results The levels of NKG2DFL and NKG2DTR in PBMC of the chronically HIV-infected patients were lower than those of the patients treated with antiretroviral therapy(P<0.01)and normal controls(P<0.01),and NKG2DFL level was significantly negatively correlated with CD4 T cell counts of chronically HIV-infected patients(r=-0.8405,P<0.01).The level changes of NKG2DFL and NKG2DTR in CD8 T cells and NK cells were consistent with PBMC,i.e.the levels of NKG2DFLand NKG2DTR in the chronically HIV-infected patients were lower than those of normal controls(P<0.05),but the levels of NKG2DFL and NKG2DTR in CD4 T cells of the chronically HIV-infected patients were instead higher than those of normal controls(P<0.05).The levels of NKG2DFL and NKG2DTRin PBMC decreased after TGF-βstimulation.Conclusion The levels of NKG2DFL and NKG2DTR in the chronically HIV-infected patients are lower than those of normal controls,especially in CD8 T and NK cells.TGF-βreduces the levels of NKG2D two splicing isoforms in PBMC.
