摘 要:目的探讨青钱柳提取物抗糖尿病大鼠氧化应激的作用。方法用链脲佐菌素(STZ)诱导SD雄性大鼠高糖血症,随机分为模型组、青钱柳提取物低、中、高剂量给药组和罗格列酮药物对照组。给药4周后(正常对照组仅正常喂食和水),按试剂盒要求测定空腹血糖值、血糖耐受力、超氧化物歧化酶(SOD)和丙二醛(MDA);荧光分光度法测定半胱氨酸蛋白酶-3(Caspase-3)含量,Western blot法测定胰腺组织中细胞色素C(cyt-C)的蛋白表达情况。结果青钱柳提取物可显著抑制STZ诱导的糖尿病大鼠的血糖升高,增强大鼠的血糖耐受力,改善SOD活性和MDA的含量、Caspase-3的水平及改变胰腺组织中的cyt-c蛋白的表达,和模型组比较差异均有统计学意义(均P<0.05)。结论青钱柳提取物具有一定的降血糖作用,其可能作用机制为抗氧化应激保护胰腺组织细胞的功能。Objective To investigate the effect of Cyclocarya paliurus extract on oxidative stress in diabetic rats.Methods Hyperglycemia SD rats induced by STZ were randomly divided into model group,high,medium,low dose of Cyclocarya paliurus extract group,and Rosiglitazone group,normal feed and water for normal control group.After 4 weeks for administration,plasma glucose,glucose tolerance,SOD,and MDA were measured;Caspase-3 concentration was assayed by fluorescence spectrophotometer,the expression of cyto chrome C(Cyt-C)in pancreatic tissue was detected by Western blot.Results Compared with the diabetic model group,plasma glucose and SOD significantly increased,while MDA in plasma and Caspase-3 significantly decreased in the Cyclocarya paliurus extract group,and the expression of Cyt-C was significantly different(all P<0.05).Conclusion The Cyclocarya paliurus extract could down regulate plasma glucose,the possible mechanism was anti-oxidative stress and the protection of the function of pancreatic tissue.
