摘 要:目的:探讨炎症因子γ-干扰素(IFN-γ)在多囊卵巢综合征(polycystic ovarian syndrome,PCOS)疾病发生及发展过程中的作用。方法:收集31例PCOS患者及22例健康志愿者肝素抗凝外周血,分离单个核细胞,分析IFN-γ+Th1和IL-4+Th2细胞比例;流式CBA法检测PCOS患者血浆IL-4、IL-6和IFN-γ水平;将人卵巢颗粒细胞瘤KGN细胞分为对照组(常规培养)和干扰素处理组(250 ng/mL IFN-γ),分析IFN-γ对KGN细胞周期、增殖及凋亡的影响。结果:与对照组相比,PCOS患者外周血单个核细胞中Th1细胞的比例明显上调(P<0.01);PCOS患者血浆中IFN-γ水平较对照组增高(P<0.05)。IFN-γ与KGN细胞体外共培养后,干扰素处理组CFSE标记KGN细胞的增殖较对照组明显右移,G0-G1期细胞增加(P<0.01),同时IFN-γ处理组凋亡比例增高(P<0.01)。结论:PCOS患者外周血中IFN-γ+Th1细胞比例和血浆中IFN-γ均增高,IFN-γ可抑制KGN细胞增殖并促进KGN细胞凋亡。Objective:To investigate the role of the inflammatory factor IFN-γin the pathogenesis and development of polycystic ovary syndrome(PCOS).Methods:The peripheral blood samples with heparin anticoagulant were collected from 31 PCOS patients and 22 healthy people.Peripheral blood mononuclear cells(PBMCs)were harvested from these samples,and the proportions of IFN-γ+Th1 and IL-4+Th2 cells in PBMCs were analyzed.The levels of IL-4,IL-6 and IFN-γin plasma of PCOS patients were detected by the flow cytometer CBA method.Human granulosa-like tumor cell line KGN cultured with IFN-γin vitro,the KGN cells were divided into two groups,control group(routine culture)and IFN-γtreated group(treated with 250 ng/mL IFN-γ).The effects of IFN-γon the cell cycle,proliferation and apoptosis of KGN cells were analyzed using flow cytometer.Results:Compared with the healty control group,the proportion of Th1 cells in PBMCs of PCOS patients was significantly up-regulated(P<0.01).The IFN-γlevel in plasma of PCOS patients was higher than that in the control group(P<0.05).After being co-cultured IFN-γwith KGN cells in vitro,the results showed that the proliferation of CFSE labeled KGN cells in the IFN-γtreated group was significantly inhibited comparing with the control group.Both the percentage of KGN cells in G0-G1 phase and the proportion of apoptotic KGN cells increased in the IFN-γtreaded group(P<0.01).Conclusion:The levels of IFN-γincreased in PCOS patients,and IFN-γinhibited the proliferation,while promoted the apoptosis of KGN cells.
