摘  要:为研究低温下去除精清的德州驴精子不同保存时间对其运动特性及母驴情期受胎率的影响,试验选择9头种公驴收集了50次射精的精液,去除精液中凝胶,离心后用INRA82稀释液重悬、稀释,获得精子密度为0.8×10^8个/mL的精液,取新鲜精液(0 h)及在5℃条件下保存24,48,72,96,120 h后的精液,使用计算机辅助精子活力分析仪检测精子的运动特性,并进行人工授精统计126头母驴情期受胎率。结果表明:与新鲜精液(0 h)相比,随着保存时间的延长,精子总活力、直线前进活力、平均运动速率及精子胞内ATP浓度逐渐降低。精液在5℃下保存时间不超过96 h时,母驴情期受胎率没有显著变化(P>0.05),而保存120 h的精液的情期受胎率显著低于0,24,48,72 h(P<0.05)。说明从精子中去除精清使驴精子免受5℃低温保存的损伤,且低温有效保存时间不超过72 h。To study the effects of different preservation time at low temperatures on sports performance and conception rate of dezhou donkey,50 ejaculations of semen from nine stud jackasses treated by removing gel were re-suspended and diluted with INRA82 diluent after centrifugation,subsequently,semen containing 0.8×10^8 sperm/mL was obtained.After 24 h,48 h,72 h,96 h or 120 h of cooled storage at 5℃,the motility characteristics of sperm and fresh sperm(0 h)were examined by a computer-aided sperm motility analyzer system,and the fetal rate of 126 female donkeys was calculated by artificial insemination.Obtained results showed that the total sperm motility,linear advancing motility,average movement rate and intracelluar ATP concentration in sperm in all groups decreased gradually with the extension of in vitro refrigeration time compared with fresh semen(0 h).When the semen was stored at 5℃for less than 96 h,there was no significant change in the pregnancy rate of the semen in the emotional stage(P>0.05),And the pregnancy rate of the semen stored for 120 h was significantly lower than that in the groups of 0 h,24 h,48 h,and 72 h(P<0.05).The results showed that semen without seminal plasma could protect donkey sperm from damage by 5℃cryopreservation,and the effective cryopreservation time was less than 72 h.
