摘  要:为了了解麝鼠PIK3cb基因及其所编码蛋白的性质功能,研究PIK3cb基因在麝鼠香腺周期变化中的作用,试验利用生物信息学分析方法对麝鼠PIK3cb基因编码蛋白的理化性质、跨膜结构、亚细胞定位、磷酸化位点、信号肽、结构域及其高级结构进行了分析,并利用RT-qPCR技术比较了PIK3cb基因在2—9月份麝鼠香腺中的转录水平差异。结果表明:PIK3cb基因的编码序列(CDS)区编码1070个氨基酸,是不稳定的亲水性蛋白;有76个磷酸化位点;该蛋白的亚细胞定位主要位于细胞质;预测该蛋白由5个结构域组成,即p85蛋白结合域(p85B)、Ras结合域(RBD)、脂质结合域C2(PI3K-C2)、PIK域(PI3Ka)和催化核心域(PI3Kc);在2—9月份的麝鼠香腺中,PIK3cb基因的表达量呈下调趋势。说明该基因在麝鼠的香腺发育或萎缩中可能起到重要调控作用。In order to understand the function of PIK3cb gene and its encoded protein in muskrat(Ondatra zibethicus Linnaeus),and to study the role of PIK3cb gene in the periodic change of muskrat scent gland,the physical and chemical properties,transmembrane structure,subcellular localization,phosphorylation site,signal peptide,domain and advanced structure of PIK3cb gene and its coding protein were analyzed by bioinformatics analysis method.In addition,RT-qPCR was used to compare the transcription levels of PIK3cb gene in muskrat scent glands from February to September.The results showed that the CDS region of PIK3cb gene encoded 1070 amino acids and was an unstable hydrophilic protein.There were 76 phosphorylation sites in this protein.The subcellular localization of the protein was mainly located in the cytoplasm.It was predicted that the protein consisted of five domains,namely,p85 protein binding domain(p85B),Ras binding domain(RBD),lipid binding domain C2(PI3K-C2),PIK domain(PI3Ka)and catalytic core domain(PI3Kc).The expression of PIK3cb gene in muskrat scent glands from February to September showed a downregulation trend.It indicated that the gene might play an important role in the development or atrophy of muskrat scent glands.
